伊立替康联合磁感应热疗对小细胞肺癌细胞增殖、凋亡及周期的影响

【摘要】 目的:探讨伊立替康联合磁感应热疗对H446细胞的作用效果。方法:研究检测磁性介质的升温性能,以及磁性介质与人小细胞肺癌H446细胞的生物相容性。将小细胞肺癌H446细胞予伊立替康、磁感应热疗及磁感应热疗联合伊立替康三种不同干预措施,采用CCK-8法检测H446细胞活力,并使用流式细胞仪检测H446细胞凋亡率及周期变化。结果:磁性介质升温性能良好且毒性小;磁感应热疗联合伊立替康20%抑制率浓度干预后,细胞抑制率为(36.58±2.10)%,细胞凋亡率为(28.90±3.64)%,均明显高于磁感应组和伊立替康组(P均<0.01);细胞周期阻滞于G2/M、S期。结论:磁感应热疗可抑制H446细胞增殖、促进凋亡,与伊立替康联合应用时具有相加抗肿瘤作用,但对细胞周期作用不明显。

【Abstract】 Objective:To investigate the effects of irinotecan combined with magnetic induction hyperthermia on H446 cells.Methods:To study and detect the heating performance of magnetic media and the biocompatibility of magnetic media with human small cell lung H446 cells.Three different interventions of small cell lung cancer H446 cells were given to irinotecan, magnetic induction hyperthermia and magnetic induction hyperthermia combined with irinotecan.CCK-8 method was used to detect the viability of H446 cells, and flow cytometry was used to detect the apoptosis rate and cycle of H446 cells variety.Results:The magnetic medium has good heating performance and low toxicity.After magnetic induction hyperthermia combined with irinotecan 20% inhibition rate concentration intervention, the cell inhibition rate was(36.58±2.10)%.The cell apoptosis rate was(28.90±3.64)%,which significantly higher than the magnetic induction group and CPT-11 group(P<0.01).Cell cycle arrest in G2/M,S phase.Conclusion:Magnetic induction hyperthermia can inhibit the proliferation of H446 cells and promote apoptosis.When combined with irinotecan, it has an additive anti-tumor effect, but has no obvious effect on the cell cycle.
