多指标综合评价丹参-红花配方颗粒提取工艺

【摘要】 目的优化丹参-红花配方颗粒的提取工艺。方法以迷迭香酸、紫草酸、丹酚酸B、羟基红花色素A为综合评价指标,采用正交试验和单因素试验,对丹参-红花配方颗粒提取有影响的乙醇浓度、料液比、提取次数、提取时间等因素进行优化;再将优化后的工艺参数用于丹参、红花的合提与单提中,筛选出最佳的提取工艺。结果以8倍量的水进行温浸提取3次,每次120 min,丹酚酸B、迷迭香酸、羟基红花黄色素A、紫草酸合提时的含量分别为59.69、2.57、5.69、7.06 mg·g-1;单提时含量分别为30.06、1.79、7.82、3.33 mg·g-1,合提优于单提。结论最佳提取工艺为:丹参-红花药对加8倍量的水浸泡60 min之后,温浸提取3次,每次120 min;该工艺稳定可行,为丹参-红花配方颗粒进一步开发应用奠定基础。

【Abstract】 Objective To optimize the extraction process of Salvia miltiorrhiza-safflower granules.Methods Taking rosmarinic acid, purpuric acid, salvianolic acid B and hydroxysafflor a as comprehensive evaluation indexes, orthogonal test and single factor test were used to optimize the factors influencing the extraction of Salvia miltiorrhiza-safflower formula granules, such as ethanol concentration, solid-liquid ratio, extraction times and extraction time; Then the optimized process parameters were used in the combined extraction and single extraction of Salvia miltiorrhiza-safflower to screen out the best extraction process.Results The contents of salvianolic acid B,rosmarinic acid, hydroxysafflor yellow A and shikonin were 59.69 mg·g-1,2.57 mg·g-1,5.69 mg·g-1 and 7.06 mg·g-1 respectively when they were extracted three times with 8 times of water for 120 min each time; The content of single extraction was 30.06 mg·g-1,1.79 mg·g-1,7.82 mg·g-1 and 3.33 mg·g-1,respectively.Conclusion The optimal extraction process is as follows: the salvia miltiorrhiza safflower pair is soaked in 8 times of water for 60 min, then extracted three times, 120 min each time; The process is stable and feasible, which lays a foundation for the further development and application of Salvia miltiorrhiza-safflower formula granules.
