HPLC法同时测定黄蜀葵花冠中芦丁、金丝桃苷和槲皮素的含量

【摘要】 目的:建立HPLC同时测定黄蜀葵花冠中芦丁、金丝桃苷和槲皮素3种活性成分的分析方法。方法:采用Waters C18色谱柱(4.6 mm×150 mm,2.7μm),乙腈-0.2%磷酸溶液作为流动相,梯度洗脱,柱温35℃,流速0.7 mL/min,进样量10μL,检测波长256 nm。结果:芦丁、金丝桃苷、槲皮素分别在0.000 5~0.010 0 mg/mL(r=0.999 95)、0.001 15~0.023 00 mg/mL(r=0.999 95)、0.003 13~0.063 00 mg/mL(r=0.999 79)范围内线性关系良好,平均加样回收率分别为96.01%(RSD=0.87%)、99.60%(RSD=0.90%)、96.92%(RSD=1.72%)。结论:本法操作简便,稳定性好,可作为黄蜀葵全花的质量控制以及同时分析黄蜀葵全花中芦丁、金丝桃苷、槲皮素的有效手段。

【Abstract】 Objective: An HPLC method for the simultaneous determination of rutin, hyperoside and quercetin from the Corolla of Abelmoschus manihot(L.) full flower was established in this paper. Methods: the Waters C18 column(4.6 mm×150 mm, 2.7 μm) was adopted to separate the samples with a gradient elution consisting of acetonitrile-0.2% phosphoric acid solution mobile phase at a flow of 0.7 mL/min. the column temperature was 35 ℃, and the detection wavelength was 256 nm, the sample volume was 10 μL. Results: The three active components showed good linear relationship respectively over the concentration range of 0.000 5~0.010 0 mg/mL(r=0.999 9)、0.001 15~0.023 00 mg/mL(r=0.999 9)、0.003 13~0.063 00 mg/mL(r=0.999 7), the average recovery was 96.01%(RSD=0.87 %)、99.60 %(RSD=0.90 %)、96.92 %(RSD=1.72 %). Conclusion: The developed method can be used as an effective means of simultaneous rutin, hyperoside and quercetin analysis.
