苦瓜籽多肽K提取工艺优化研究

【摘要】 苦瓜籽多肽K能有效改善胰岛素抵抗状态,具有显著的降糖效果。为提高苦瓜籽中多肽K的提取率,选育出多肽K含量较高的苦瓜籽品种,以5个品种的苦瓜籽为原料,采用单因素试验优化碱提酸沉法提取苦瓜籽中多肽K的工艺条件。结果表明:苦瓜籽中多肽K的最佳提取工艺条件为料液比1∶20(g/mL)、碱液pH 9.5、提取时间60 min、提取温度50℃、酸沉pH 3.0。不同品种苦瓜籽的多肽K含量差异显著,筛选出2个含量相对较高的品种,分别是桂农科9号苦瓜籽和绿水果苦瓜籽。其中桂农科9号苦瓜籽的多肽K含量最高,为2.06 mg/g,经碱提酸沉工艺优化提取后多肽K提取率为0.18 mg/g。

【Abstract】 Hypoglycemic polypeptide-K of bitter gourd seeds was to increase sensitivity of tissue to insulin and improve insulinresistance,which was remarkable hypoglycemic effect. In order to improve the extraction rate of polypeptide-K from bitter gourd seeds,breed varieties with high content of polypeptide K. In this study,five varieties of bitter gourd seeds were used materials,the single factor test optimization combined with alkali extraction-acid precipitation was used to extract the polypeptide-K. The results showed that alkali extractionacid precipitation method was used to extract the polypeptide-K. The optimum technological conditions were liquid ratio 1 ∶ 20(g/mL),alkaline solution pH 9.5,temperature 50 ℃,extraction time 60 min,acid precipitation pH 3.0. The extraction ratio of polypeptide K in bitter gourd seeds varieties was significantly different. The high content of polypeptide-K were Guinongke 9 and Green fruit bitter gourd seeds,and Guinongke 9 was the highest,which was 2.06 mg/g. After optimization of alkali extraction and acid precipitation process,the extraction of polypeptide K was 0.18 mg/g.
