伏马毒素B1对小鼠血液学指标及凋亡的影响

【摘要】 为探讨伏马毒素B1(FB1)对小鼠血液学指标及细胞凋亡的影响,试验选用昆明小鼠饲喂含FB1的饲料,连续45 d后称量体重并计算肾脏、脾脏和肝脏脏体比,测定血常规和血液生化指标,采用Tunel法检测脾脏细胞凋亡的变化,qPCR测定肝脏、脾脏和肾脏中凋亡相关基因的mRNA表达量。结果表明:浓度为36.82 mg/kg的FB1染毒小鼠45 d后,小鼠精神萎靡、食欲下降、体重降低,脾体比显著下降0.1%(P <0.05)。与对照组相比,血液中淋巴细胞减少,总蛋白(TP)和白蛋白(ALB)的含量分别降低约11.32 g/L和5.99 g/L(P <0.05),尿素(Urea)的含量明显上升约4.14 mmol/L(P <0.05),天冬氨酸氨基转移酶(AST)、丙氨酸氨基转移酶(ALT)的酶活力显著升高(P <0.05),分别升高约51.93 U/L和38.94 U/L。脾脏中细胞凋亡的数量明显增加,且肝脏和脾脏中Bax、Caspase 3、Caspase 9和P53的表达都显著升高(P <0.05),脾脏中Fas的表达显著上升(P <0.05),肾脏中Bax的表达显著上升(P <0.05)。综上所述,FB1能够影响小鼠的生长状态和血液学指标,促使肝、脾和肾脏损伤。

【Abstract】 To investigate the effects of fumonisin B1(FB1) on hematological indexes and cell apoptosis of mice,Kunming mice were selected in the experiment and fed with FB1 contaminated feed.After 45 consecutive days feeding,the body weight was measured and the ratio of kidney,spleen and liver were calculated,while blood routine and blood biochemical indicators were measured. The Tunel method was used to detect the changes cell apoptosis in spleen,and real-time fluorescence quantitative PCR was used to determine the mRNA expression of apoptosis-related genes in the liver,spleen and kidney. The results showed that after exposed to FB1 at a concentration of 36.82 mg/kg for 45 days,the mice were depressed in spirit,decreased appetite and weight,and the spleen ratio was decreased significantly by 0.1%(P < 0.05).Compared with the control group,the concentration of lymphocytes were decreased,and the contents of total protein(TP) and albumin(ALB) decreased by about 11.32 g/L and 5.99 g/L(P < 0.05),and the content of urea was about significantly increased by 4.14 mmol/L(P < 0.05),while aspartate transaminase(AST)and alanine aminotransfease(ALT) activity were significantly increased by about 51.93 U/L and 38.94 U/L respectively(P < 0.05).The number of apoptosis was significantly increased in spleen.The expressions of Bcl-2 associated X protein(Bax),cysteine-dependent aspartate-specifc proteases 3(Caspase 3),Caspase 9 and P53 in spleen and liver were significantly increased(P < 0.05). The expression of Fas in the spleen increased significantly(P < 0.05),and the expression of Bax in the kidney increased significantly(P <0.05). The results indicated that FB1 could affect the growth state and hematological indicators of mice,and promote liver,spleen and kidney injury.
