夹脊电针对脊髓损伤大鼠运动功能、神经肌肉形态学影响及机制研究
Study on the Effect and Mechanism of Jiaji Electroacupuncture on Motor Function, Neuromuscular Morphology of Spinal Cord Injury Rats
【摘要】 目的研究夹脊电针对脊髓损伤(Spinal cord injury, SCI)大鼠运动功能、神经肌肉形态学的影响,并探讨相关机制。方法将40只Wistar成年雌性大鼠随机分为假手术组、模型组、电针组、阳性对照组。假手术组仅做椎板切除不造成SCI,其余各组采用Allen′s改良式重物撞击法建立SCI模型。假手术组及模型组不予以治疗,电针组以夹脊电针治疗,阳性对照组以注射用单唾液酸四己糖神经节苷脂钠治疗。以Basso-Beattie-Bresnahan(BBB)评分量表评价大鼠运动功能,光镜与电镜下观察大鼠脊髓与腓肠肌形态变化,计算肌纤维横截面积和直径,Western blotting检测大鼠脊髓组织中B淋巴细胞瘤-2(Bcl-2)、Bcl-2 Assaciated X的蛋白(Bax)表达情况。结果与假手术组比较,模型组BBB评分术后7 d、14 d、28 d的BBB评分更低,神经元有明显的变性、坏死及炎性浸润,髓鞘分离,腓肠肌肌纤维萎缩,肌纤维横截面积与直径更小,Bcl-2相对表达量降低,Bax相对表达量与Bax/Bcl-2更高,差异有统计学意义(P<0.05)。与模型组比较,电针组术后7 d、14 d、28 d的BBB评分更高,神经元损害减轻,少部分髓鞘轴索分离,存在肌纤维萎缩,肌纤维横截面积与直径更大,Bcl-2相对表达量更高,Bax相对表达量与Bax/Bcl-2更低,差异有统计学意义(P<0.05)。与电针组比较,阳性对照组术后7 d、14 d、28 d的BBB评分更高,神经元损害、髓鞘轴索分离与肌纤维萎缩减轻,肌纤维横截面积与直径更大,Bcl-2相对表达量更高,Bax相对表达量与Bax/Bcl-2更低,差异有统计学意义(P<0.05)。结论对脊髓损伤大鼠采取夹脊电针治疗可改善大鼠的运动功能,促进神经元及肌纤维形态的恢复,其机制可能与调节Bcl-2、Bax的表达有关。
【Abstract】 Objective To study the effects of Jiaji electroacupuncture on motor function and neuromuscular morphology of spinal cord injury(SCI)rats, and to explore related mechanisms.Methods Forty adult female Wister rats were randomly divided into sham operation group, model group, electroacupuncture group, and positive control group.In the sham operation group, only laminectomy did not cause SCI,and the other groups used Allen′s modified weight impact method to establish SCI models.The sham operation group and the model group were not treated, the electro-acupuncture group was treated with Jiaji electro-acupuncture, and the positive control group was treated with injection of monosialotetrahexose ganglioside sodium.The motor function of rats was evaluated by the Basso-Beattie-Bresnahan(BBB)scoring scale.The morphological changes of the rat spinal cord and gastrocnemius were observed under light and electron microscopes, and thecross-sectional area and diameter of muscle fibers were calculated.Western blotting was used to detect the expression of B lymphoma-2 gene(Bcl-2)and Bcl-2 Assaciated X protein(Bax)in rat spinal cord tissue.Results Compared with the sham operation group, the BBB scores of the model group were lower on the 7 d, 14 d, and 28 d days after surgery, the neurons had obvious degeneration, necrosis and inflammatory infiltration, myelin sheath separation, gastrocnemius muscle fiber atrophy, the cross-sectional area and diameter of muscle fiber were smaller, the relative expression of Bcl-2 was lower, and the relative expression of Bax was higher than that of Bax/Bcl-2,and the difference was statistically significant(P<0.05).Compared with the model group, the BBB scores of the electroacupuncture group were higher on the 7 d, 14 d, and 28 d days after surgery, the neuron damage was reduced, a small part of the myelin sheath was separated, there was muscle fiber atrophy, and the muscle fiber cross-sectional area and diameter were larger, the relative expression of Bcl-2 was higher, and the relative expression of Bax was lower than that of Bax/Bcl-2,and the difference was statistically significant(P<0.05).Compared with the electroacupuncture group, the positive control group had higher BBB scores on the 7 d, 14 d, and 28 d days after surgery, the neuronal damage, myelin axon separation and muscle fiber atrophy were reduced, the cross-sectional area and diameter of muscle fibers were larger, the relative expression of Bcl-2 was higher, and the relative expression of Bax was lower than that of Bax/Bcl-2,and the difference is statistically significant(P<0.05).Conclusion Jiaji electroacupuncture can improve the motor function and promote the morphological recovery of neurons and muscle fibers in rats with spinal cord injury.The mechanism may be related to the regulation of the expression of Bcl-2 and Bax.
