树莓成熟胚组织培养技术研究
Study on tissue culture technology of mature embryo of raspberry
【摘要】 以不同品种树莓杂交种子为材料,采用成熟胚组织培养技术,研究了不同植物生长调节剂组合对其发芽、幼苗增殖和生根的影响。结果表明:树莓杂交种子(HT)成熟胚最适灭菌条件为0.1%Hg Cl2灭菌10 min;在1∕2MS+6-BA 0.5 mg∕L+IBA(1.5—2.0)mg∕L培养基上,树莓杂交种子萌发率最高达88.9%(RQ);无菌苗在MS+6-BA(0.5—0.8)mg∕L+IBA 0.1 mg∕L+GA30.5 mg∕L培养基上的增殖倍数最高达4.87倍(AT),且植株生长健壮;在生根培养基1∕2MS+IBA(0.2—0.3)mg∕L培养时生根率最高达96.35%(WH)。可见,6-BA、IBA和GA3对树莓种子萌发和成苗具有明显促进作用,优化的培养基可显著提高树莓杂交种子成熟胚的萌发、成苗和生根。
【Abstract】 The effects of different combinations of plant growth regulators on germination,seedling proliferation and rooting of hybrid seeds of different raspberry varieties were studied by mature embryo tissue culture.The results showed that the optimum sterilization condition for mature embryos of raspberry hybrid seeds(HT) was 0.1% HgCl2 sterilization for 10 min;The highest germination rate of raspberry hybrid seeds was 88.9%(RQ)on 1/2 MS+6-BA 0.5 mg/L+IBA(1.5-2.0) mg/L medium;The multiplication of sterile seedlings on MS+6-BA(0.5-0.8) mg/L+IBA 0.1 mg/L+GA3 0.5 mg/L medium was up to 4.87 times(AT),and the plants grew robustly;When the rooting medium was 1/2 MS+IBA(0.2-0.3) mg/L,the rooting rate was 96.35%(WH).It could be seen that 6-BA,IB A and GA3 could significantly promote the germination and seedling establishment of raspberry seeds.The optimized medium could significantly improve the germination,seedling establishment and rooting of mature embryos of raspberry hybrid seeds.
