鸡蛋溶菌酶单克隆抗体的制备与鉴定
Preparation and characterization of monoclonal antibody against egg lysozyme
【摘要】 为建立一种便捷的鸡蛋溶菌酶定量测定方法,将鸡蛋溶菌酶免疫BALB∕c小鼠,取免疫小鼠脾细胞与SP2∕0骨髓瘤细胞融合,用ELISA方法筛选阳性杂交瘤细胞,制备腹水并纯化,用间接ELISA和Western blot试验方法检测纯化抗体的特异性。结果表明:通过筛选获得1株稳定分泌特异性抗体的杂交瘤细胞系3E4,其抗体亚型为Ig G2b,细胞培养上清和腹水效价分别达2 560和1.0×106;MAb的特异性鉴定结果显示,该株细胞系所产生的抗体能够特异地与鸡蛋溶菌酶蛋白反应。该抗体的制备为鸡蛋溶菌酶检测方面的研究提供了基础。
【Abstract】 In order to establish a convenient method for the quantitative determination of lysozyme from egg white,the lysozyme was immunized into BALB∕c mice,and the spleen cells from immunized mice were fused with SP2∕0 hybridoma cells,and the hybridomas secreting monoclonal antibody were established and detected by ELISA and Western blotting.The results showed that 3E4 monoclonal antibody(MAb)was obtained with Ig G2b subtype.The titers of supernatant and ascites were 2 560 and 1.0×106,respectively.The MAb reacted specifically with lysozyme from chicken egg white.The preparation of this MAb would facilitate the study of lysozyme.
