首发精神分裂症DRD2 C957T多态性及认知功能的相关性研究
Polymorphism and correlation with cognitive function of dopamine receptor D2(DRD2) C957T in patients with first-episode schizophrenia
【摘要】 目的探讨DRD2 C957T多态性在首发精神分裂症患者与健康人群中分布的差异,以及与认知功能的相关性。方法采用横断面研究,收集首发精神分裂症患者74例(研究组)和健康对照73名(对照组),应用PCR直接测序法测定C957T基因型,使用MATRICS共识认知成套测验(MCCB)评估认知功能。结果 C957T位点在两组之间的基因型和等位基因频率比较,差异均具有统计学意义(P <0.05)。进一步对性别分组,C957T位点在两组男性中基因型和等位基因频率比较,差异均具有统计学意义(P <0.05)。研究组MCCB中TMT、HVLT-R、BVMT-R、CPT-IP及Stroop色词测验评分均低于对照组(P <0.05)。研究组中,CC基因型BVMT-R分测验评分高于CT基因型(P <0.05)。TMT、HVLT-R、BVMT-R和CPT-IP评分与精神分裂症患病风险呈负相关(P <0.05)。结论 DRD2 C957T可能与男性精神分裂症发病相关,精神分裂症患者认知功能受损,携带不同基因型患者与简易视觉功能受损可能存在关联。
【Abstract】 Objective To explore the distribution differences of dopamine receptor D2( DRD2) C957 T polymorphism in patients with first-episode schizophrenia and healthy controls and its correlation with cognitive function. Methods A crosssectional study was conducted in 74 patients with first-episode schizophrenia( study group) and 73 normal controls( control group). C957 T genotypes were determined by PCR direct sequencing and cognitive function was evaluated by MATRICS Consensus Cognitive Battery( MCCB). Results There existed significant differences in distribution frequencies of genotypes and alleles of C957 T between two groups as well as between males of two groups( P < 0. 05). The subscale scores of TMT,HVLT-R,BVMT-R,CPT-IP and Stroop Color-Word Test of MCCB in study group were significantly lower than those in control group( P <0. 05). In study group,the subscale score of BVMT-R in patients with CC genotype was significantly higher than that in patients with CT genotype( P < 0. 05). The subscale scores of TMT,HVLT-R,BVMT-R,CPT-IP were negatively correlated with the risk of schizophrenia( P < 0. 05). Conclusion The DRD2 C957 T may be related to the onset of schizophrenia in males,and the cognitive function in schizophrenics is impaired. Patients with different genotypes may be related to the impairment of simple visual function.
