柴胡茎叶总黄酮生物活性研究
Study on the biological activity of total flavonoids from the stems and leaves of Bupleurum chinense
【摘要】 目的研究柴胡茎叶总黄酮提取方法及生物活性。方法以总黄酮提取率和浸膏得率为指标确定柴胡茎叶总黄酮的提取方法。在此基础上制备柴胡茎叶总黄酮水溶液,研究柴胡茎叶中总黄酮抗疲劳作用。以耐力游泳小鼠作为抗疲劳动物模型,随机分为阴性对照组、阳性对照组和3个不同剂量给药组(0.5、1.0和1.5g·mL-1),每组各10只。通过对肝糖原、肌糖原、乳酸脱氢酶(LDH)、血乳酸(LAC)和血尿素氮(BUN)、超氧化物歧化酶(SOD)、负重游泳时间等指标研究柴胡茎叶总黄酮抗疲劳作用。结论柴胡茎叶总黄酮具有抗疲劳作用,其作用机理主要是通过运动肌糖供给实现抗疲劳作用。
【Abstract】 Objective To study the extraction method and biological activity of total flavonoids from Bupleurum stems and leaves. Methods The method of extracting total flavonoids from Bupleurum stems and leaves was determined by the extraction rate of total flavonoids and the yield of extract. Basing on that, an aqueous solution of total flavonoids from Bupleurum stems and leaves was prepared to study its antifatigue effect. Using endurance swimming mice as the anti-fatigue animal model, they were randomly divided into a negative control group, a positive control group, and 3 different dose groups(0.5, 1.0 and 1.5g·mL-1), with 10 mice in each group. The anti-fatigue effects of total flavonoids in stems and leaves of Bupleurum were studied by mensurating liver glycogen, muscle glycogen, lactate dehydrogenase(LDH),blood lactic acid(LAC), blood urea nitrogen(BUN), superoxide dismutase(SOD), exhaustive swimming time and other indicators. Conclusion Total flavonoids from Bupleurum stems and leaves have anti-fatigue effects, the mechanism of which is mainly through ensuring the motor system muscle glycogen.
