新癀片外用给药对小鼠镇痛模型的有效成分研究
Study on effective components of analgesic model of mice by external administration of Xinhuang tablets
【摘要】 目的分析新癀片外用对小鼠镇痛模型的有效成分,为新癀片镇痛的药效成分研究提供科学依据和参考。方法 ICR小鼠50只,雌雄各半,随机分为5组:模型组、阳性对照组、给药组、全方中等剂量吲哚美辛组、全方缺吲哚美辛群药组,每组10只。连续给药3 d,给药后40 min醋酸造模,立即观察并记录小鼠出现扭体的潜伏期和20 min内小鼠发生扭体反应的次数。雌性ICR小鼠50只,根据基础痛阈值分为5组,给药后30 min、60 min、120 min、180 min和240 min测定痛阈值。结果与模型组相比,阳性对照组、给药组和全方中等剂量吲哚美辛组的潜伏期均延长(P <0.05),扭体次数降低(P <0.05)。而全方缺吲哚美辛群药组潜伏期较模型组有所延长,扭体次数有所减少,但无统计学差异(P> 0.05)。给药后30 min到给药后240 min,阳性对照组和给药组的痛阈值较模型组有所延长(P <0.05)。而全方中等剂量吲哚美辛组和全方缺吲哚美辛群药组的痛阈值与模型组比较差异无统计学意义(P>0.05)。结论新癀片中的吲哚美辛和中药成分之间相互作用,相互协同,共同参与小鼠的镇痛作用。
【Abstract】 Objective To analyze the effective components of Xinhuang tablets for external use on the analgesic model of mice,which would provide scientific basis and reference for the analgesic components of Xinhuang tablet. Methods50 ICR mice,half male and half female,and they were randomly divided into 5 groups: model group,positive control group,administration group,isodose indomethacin group,deficiency indomethacin group,10 mice in each group. The mice were given for 3 days continuously,acetic acid was used for modeling 40 min after administration,the incubation period of mice writhing and the times of mice writhing reaction within 20 min were observed and recorded immediately. 50 female ICR mice were divided into the above 5 groups according to the basic pain threshold,pain thresholds were measured 30 min,60 min,120 min,180 min and 240 min after administration. Results Compared with the model group,the incubation period of the positive control group,the administration group and the isodose indomethacin group were prolonged( P < 0. 05),and the number of writhings decreased( P < 0. 05). However,in the deficiency indomethacin group,the incubation period was prolonged and the number of writhings decreased,but the difference was not statistically significant( P > 0. 05). From 30 min to 240 min after the administration,the pain threshold of the positive control group and the administration group was longer than that of the model group( P < 0. 05). However,there was no significant difference in pain threshold between the isodose indomethacin group and deficiency indomethacin group of the whole prescription and the model group( P > 0. 05). Conclusion The indomethacin and the components of traditional Chinese medicine in Xinhuang tablets are interacted and cooperated with each other,to participate in the analgesic effect of mice.
