乳腺癌细胞中的基因转录暂停现象研究

【摘要】 为揭示基因转录暂停机制在乳腺癌发生中的作用,利用生物信息学方法鉴定了人乳腺上皮细胞和6种乳腺癌上皮细胞中的转录暂停基因,分析了转录暂停基因的表达变化和管家基因的暂停分布。结果表明:1)癌细胞中暂停基因占比较高,基因转录暂停程度与其表达量呈负相关。2)转录暂停的基因倾向于在核糖核蛋白复合物生物合成和RNA剪接等通路中富集,癌细胞中的管家基因同样发现有大量活跃的转录暂停基因。3)暂停位置附近显著富集KLF5、NRF1、ELK4 3种转录因子的结合位点。上述研究为认识细胞癌变的发生提供了基因转录暂停的全组学分子基础,有助于全面和深入认识癌症发生的分子机制。

【Abstract】 In order to reveal how transcriptional pause regulate the occurrence of breast cancer, this study used bioinformatics methods to identify transcriptional pause genes, analyze the expression changes of transcription pause genes and the pause distribution of housekeeping genes in the human mammary epithelial cell and six breast cancer epithelial cells. The results showed: 1) The proportion of pause genes in cancer cells is high, and the degree of gene transcription pause is negatively correlated with its expression. 2) Genes with transcriptional pauses tend to be enriched in pathways such as ribonucleoprotein complex biosynthesis and RNA splicing. A large number of active transcription pause genes is also found in house keeping cancer cells. 3) The binding sites of KLF5, NRF1 and ELK4 transcription factors are significantly enriched near the pause position. The above studies provide a genome-wide molecular basis for understanding the occurrence of cell carcinogenesis, and is helpful for a comprehensive and in-depth understanding of the molecular mechanism of cancer.
