原位电离质谱法表征及成像橙皮中多甲氧基黄酮类化合物

【摘要】 多氧基黄酮(Polymethoxy flavonoids,PMFs)一类具有抗炎、抗菌、抗癌功效的天然黄酮类化合物,在鲜橙内含量丰富,因此通过2种直接质谱分析手段——内部萃取电喷雾电离质谱法(i EESI MS)和表面解析电喷雾电离质谱成像(DESI MSI)对橙皮中的PMFs进行了表征。i EESI MS结合串联质谱功能测定橙皮不同部位黄酮类化合物的种类;DESI MSI对鲜橙果皮横截面的PMFs进行成像。结果显示,通过对i EESI信号进行串联质谱分析,橙皮油胞层含有大量的川陈皮素、橙皮素及四甲氧基黄酮,质谱成像结果表明,物质仅在橙皮油胞层分布,与i EESI检测结果一致,为废弃橙皮利用提供新思路。

【Abstract】 Polymethoxy flavonoids(PMFs),a class of natural flavonoids with anti-inflammatory,antibacterial and anticancer properties. In this study,PMFs in fresh orange peel were characterized by two methods of direct mass spectrometry——Internal extraction electrospray ionization mass spectrometry(i EESI MS) and desorption electrosprayionization mass spectrometry images(DESI MSI). The classification of flavonoids in different parts of orange peel was determined by i EESI MS combined with tandem mass spectrometry. DESI MSI imaged PMFs in cross section of fresh orange peel. The results showed that by tandem mass spectrometry analysis of i EESI MS,the cell layer of orange peel oil contained a large amount of Tangerine,hesperidin and tetramethoxylflavone,and the DESI MSI results showed that the substances were only distributed in the cell layer of orange peel oil,which was consistent with the detection results of i EESI. The purpose of this study was to provide new ideas for the utilization of discarded orange peel.
