LED光质对两种金线莲组培苗生理特征和代谢物质的影响

【摘要】 为明确光质对金线莲组培苗生长和代谢物质积累的影响,本文以福建金线莲(Anoectochilus roxburghii)和台湾金线莲(A.formosanus)组培苗为实验材料,在红蓝光2:1(R2B1)、红蓝光1:2(R1B2)和白光(W)3种LED光处理下培养60 d,采用分光光度法测定叶绿素、可溶性糖、可溶性蛋白、总酚和总黄酮含量,利用高效液相色谱法结合标准品测定槲皮素、异鼠李素、山奈酚、阿魏酸和绿原酸含量。结果表明:两种金线莲在R2B1下株高、叶长、叶宽、鲜重、叶绿素和5种酚酸类化合物含量均达到最高,在R1B2下可溶性蛋白含量均达到最低。R2B1下福建金线莲总酚含量(3.33±0.22)mg·g-1显著高于白光,而R1B2下台湾金线莲可溶性糖(4.06±0.22)mg·g-1和总黄酮含量(1.37±0.07)mg·g-1显著高于白光。R2B1下福建金线莲中槲皮素含量和异鼠李素含量分别达到(36.52±0.91)和(61.35±2.45)μg.g-1,台湾金线莲中绿原酸含量达到(40.09±3.79)μg.g-1。R2B1可以作为金线莲组织培养的适宜光配方,为LED在金线莲组培方面的应用提供科学依据。

【Abstract】 To investigate the effect of the red/blue light quality ratio on the growth and metabolites accumulations of A.roxburghii and A.formosanus,the tissue culture plants of A.roxburghii and A.formosanus were treated with three LED lights of R2 B1(with Red/Blue ratio of 2:1),R1 B2(with Red/Blue ratio of 1:2),and W(white light).After 60 d of illumination, the content of chlorophyll, soluble sugar, soluble protein, total phenols, and total flavonoids were determined by spectrophotometry, and the content of quercetin, isorhamnetin, kaempferol, ferulic acid, and chlorogenic acid were determined by HPLC combined with standards.Under the R2 B1 light, the fresh weight, dry weight, contents of chlorophyll and phenolic acid compounds of the two species reached the highest.The soluble protein content of the two species under the R1 B2 light reached the lowest.Under the R2 B1 light, the total phenol content of A.roxburghii was significantly higher than that of W.But under the R1 B2 light, the content of soluble sugar and total flavonoids of A.formosanus was significantly higher than that of W.Under the R2 B1 light, the contents of quercetin and the isorhamnetin in A.roxburghii were(36.52±0.91)and(61.35±2.45) μg.g-1,respectively, and the content of chlorogenic acid in A.formosanus was(40.09±3.79) μg.g-1.Therefore, the R2 B1 compound light can be used as a suitable light formula for A.roxburghii and A.formosanus,and it can provide scientific basis for the application of LED in the tissue culture of A.roxburghii and A.formosanus.
