通过TCGA数据库研究干扰素调节因子7在肾透明细胞癌中的作用

【摘要】 目的通过TCGA数据库研究干扰素调节因子7 (interferon regulatory factor 7,IRF7)在肾透明细胞癌(clear cell renal cell carcinoma,ccRCC)中的表达,并分析其表达水平对ccRCC患者预后的影响,探讨与其表达相关的基因网络。方法利用Ualcan、GEPIA、人类蛋白质图谱(human protein atls,HPA)、TIMER 2.0等数据库分析ccRCC组织中IRF7 mRNA和蛋白质水平变化。运用Ualcan、GEPIA、HPA数据库研究IRF7表达与ccRCC临床病理学参数的关系及对预后的影响。在String-DB数据库中探索并验证IRF7基因在细胞信号转导通路中的位置以及与其关系密切的上下游基因。结果 IRF7在ccRCC中的表达升高,差异有统计学意义(P<0.01),且与ccRCC的不良预后相关,差异有统计学意义(P<0.05),其表达与ccRCC的临床分期、有无淋巴结转移及ccRCC亚型相关;预后分析显示,IRF7高表达组患者较低表达组生存率降低,差异有统计学意义(P<0.01)。结论通过数据库信息挖掘发现,IRF7在ccRCC组织中呈高表达,且高表达是ccRCC患者预后的不利因素,为后续的ccRCC研究提供重要的理论依据。

【Abstract】 Objective This study aimed to examine the expression of interferon regulatory factor 7(IRF7) in clear cell renal cell carcinoma(ccRCC) according to the TCGA database, and by analyzing the effects of its expression level on the prognosis of patients with ccRCC, and exploring the gene networks related to its expression. Methods Changes in IRF7 mRNA and protein levels in ccRCC tissues were analyzed with database tools such as ualcan, gepia, HPA and timer 2.0. The relationship between IRF7 expression and clinicopathological parameters of ccRCC and its effects on prognosis were studied by using the ualcan, gepia and HPA databases. We explored and verified the position of the IRF7 gene in the cell signal transduction pathways and its closely related upstream and downstream genes in the String DB database. Results The expression of IRF7 was elevated in ccRCC(P<0.01) and was associated with poor ccRCC prognosis. Its expression was associated with the clinical stage, lymph node metastasis and subtype of ccRCC. Prognostic analysis showed that the survival rate of patients with high expression of IRF7 was lower than that of patients with low expression of IRF7(P<0.01). Conclusion Through database information mining, we found that IRF7 is highly expressed in ccRCC, and this high expression is associated with poor prognosis in patients with ccRCC. These findings provide an important theoretical basis for future studies of ccRCC.
