风柜斗草醇提物对小鼠酒精性肝损伤的保护作用

【摘要】 研究风柜斗草醇提物对小鼠急性酒精性肝损伤的保护作用并探讨其可能机制。实验选择60只体重均衡的ICR雄鼠(10只/组),设置空白组、模型组、风柜斗草醇提物剂量组(200、400、800 mg/kg)和联苯双酯阳性对照组(200 mg/kg)。经处理后,检测肝组织中的超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-PX)、丙二醛(MDA)的水平及血清中丙氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST)的活性,并进行肝脏HE染色观察组织病理学病变。结果表明:风柜斗草醇提物能显著降低血清中ALT、AST活性,提高肝组织中SOD及GSH-PX活性,降低肝组织中MDA水平;病理学观察发现风柜斗草醇提物能减少肝脏脂肪变性及降低肝细胞的损伤。风柜斗草醇提物对乙醇所致的小鼠急性酒精性肝损伤具有保护作用。

【Abstract】 To study the protective effect of ethanol extract from Sarcopyramis nepalensis wall on acute alcoholic liver injury in mice, and to explore its possible mechanism, 60 ICR male mice were randomly divided into six groups(10 mice/group), namely the blank group, the model group, the dose group(200, 400, 800 mg/kg) and the positive control group(200 mg/kg). At the end of the experiment, kits were used to assay liver superoxide dismutase(SOD) and glutathione peroxidase(GSH-PX) activity, and serum alanine aminotransferase(ALT) and aspartate aminotransferase(AST) activity. Furthermore, the liver HE staining was conducted to observe the histopathological lesions. The results showed that the alcohol extract from Sarcopyramis nepalensis wall can significantly reduce the activity of serum ALT and AST, improve the activity of liver SOD and GSH-PX, and also reduce the group of MDA. The results of HE staining showed that Sarcopyramis nepalensis wall extract improved hepatic steatosis and liver cell damage. The conclusion is that the ethanol extract from Sarcopyramis nepalensis wall exerts protective effect on acute alcoholic liver injury, and this effect may be due to its anti-oxidation activity.
