9种中药单体的体外抗单梗着色霉活性评价
Antifungal effects of 9 Chinese herbal monomers against Fonsecaea monophora in vitro
【摘要】 目的:体外检测中药单体对单梗着色霉的抑菌活性,为临床采用中药治疗着色芽生菌病提供实验依据。方法:11株单梗着色霉菌株为实验菌株,包括1株标准株、10株临床分离株。参照CLSI制定的M38-A2,采用微量稀释法检测蛇床子素、姜黄素、白鲜碱、知母皂苷、黄芩苷、苦参碱、丹皮酚、斑蝥素和大黄酚9种中药单体对单梗着色霉的抑制作用。结果:9种中药单体最小抑菌浓度(MIC)的几何均数为14.11~272.65μg/mL,蛇床子素、姜黄素和白鲜碱的抑菌活性最佳,几何均数分别为14.11、28.21、52.98μg/mL;蛇床子素的抗菌作用最强,MIC范围为8~32μg/mL。9种中药单体最小杀菌浓度(MFC)为128~>1 024μg/mL;蛇床子素的MFC值最低,几何均数为272.65μg/mL。结论:蛇床子素、姜黄素、白鲜碱、知母皂苷、黄芩苷、苦参碱、丹皮酚、斑蝥素和大黄酚对单梗着色霉具有一定的抑菌活性,不同中药单体抑菌活性存在差异。
【Abstract】 Objective:To evaluate antifungal effects of 9 Chinese herbal monomers against Fonsecaea monophora in vitro, providing an experimental evidence for the clinical treatment of chromoblastomycosis with Chinese medicine. Methods: There were 11 strains of Fonsecaea monophora, including 1 standard strain and 10 clinical isolates. According to the M38-A2 protocol formulated by CLSI, the micro-liquid-based dilution method was used to detect the antifungal effects of 9 Chinese herbal monomers against 11 strains of Fonsecaea monophora. Result:The MIC value of the quality control plant was within the reference range, and the positive control grew well. The geometric mean range of the minimum inhibitory concentration(MIC) of 9 kinds of traditional Chinese herbal monomers was 14.11~272.65 μg/mL. The effect of osthole, curcumin and dictamnine were the best, and their geometric mean were 14.11 μg/mL, 28.21 μg/mL and 52.98 μg/mL, respectively. Osthole had the strongest antifungal effect on Fonsecaea monophora, showing the range of MIC was 8~32 μg/mL. The minimum fungicidal concentration(MFC) ranged from 128 μg/mL to >1 024 μg/mL. Osthole had the lowest MFC value, and GM MFC was 272.65 μg/mL.Conclusion:Osthole, curcumin, dictamnine, anemarrhena saponins, baicalin, matrine, paeonol, cantharidin and chrysophanol exhibits antifungal effects against Fonsecaea monophora.
