急性冷暴露对仔猪多器官miR-383表达的影响
Effect of acute cold exposure on expression of miR-383 in multiple organs of piglets
【摘要】 试验旨在探讨急性冷暴露对仔猪体内miR-383表达量的影响。试验选取16头30日龄断奶仔猪,分为对照组和冷暴露组,冷暴露条件为(4±2)℃。冷暴露组又分为3个处理,分别进行2、6、12 h冷暴露,每组4头仔猪。采用ELISA法检测血清中皮质醇(CORT)含量;次用qRT-PCR法检测肺、小肠、肌肉、肾、脾和肝中miR-383的表达水平。结果显示,冷暴露6 h后,仔猪血清CORT含量显著升高(P<0.01);各器官miR-383表达水平均出现显著变化(P<0.05),但变化出现的时间点和表现出的趋势不同。研究表明,4℃冷暴露6 h可使仔猪处于急性冷应激状态;从miR-383表达量角度分析,肺、肌肉、肝和脾对冷刺激响应迅速,小肠和肾的响应时间较长;急性冷暴露可使仔猪肝脏中miR-383表达量持续显著上升(P<0.05)。
【Abstract】 The purpose of the experiment was to investigate the effect of acute cold exposure on the expression of miR-383 in piglets. Sixteen 30-day-old weaned piglets were selected for the experiment, and they were divided into a control group and a cold exposure group. The cold exposure condition was(4±2) ℃. The cold-exposure group was divided into three treatments, which were cold-exposed for 2, 6, and 12 h, with four heads in each group. ELISA method was used to detect serum cortisol(CORT) content,and qRT-PCR method was used to detect the expression level of miR-383 in lung, small intestine, muscle, kidney, spleen and liver. The results showed that after 6 h of cold exposure, the serum CORT content of piglets increased significantly(P<0.01); the expression level of miR-383 in various organs changed significantly(P<0.05), but the time point and trend of the change appeared different. The study indicates that 4 ℃cold exposure for 6 h can make piglets in an acute cold stress state. From the perspective of miR-383 expression, lung,muscle, liver and spleen respond quickly to cold stimulation, and the small intestine and kidney have a longer response time. Acute cold exposure can continuously increase the expression of miR-383 in piglet liver(P<0.05).
