凝血酶敏感蛋白-1与血管新生
Thrombospodin-1 and angiogenesis
【摘要】 凝血酶敏感蛋白-1(Thrombin-sensitive protein,Thrombospodin-1,TSP-1)是在人体中首先被发现的、由3条分子量约为145kDa的相同肽链组成的同源三聚体,其多个片段与血管新生以及细胞迁移、黏附、侵袭、凋亡等过程有关.TSP-1的结构域可与多种细胞表面受体分子相互作用,因而具有多种功能.TSP-1通过抑制生长因子与受体结合,或与血管内皮细胞表面多种黏附分子结合抑制肿瘤的血管新生,进而控制肿瘤的发生与发展,也有研究证明来源于TSP-1的多肽片段同样具有抗血管新生和抗肿瘤功能.对人源TSP-1的结构、受体及抗血管新生功能进行了综述,以期为以TSP-1为基础的血管新生抑制剂研究提供参考.
【Abstract】 Thrombospodin-1(TSP-1)is a homotrimer first discovered in the human body and composed of three identical peptide chains with a molecular weight of approximately 145 kDa.Its multiple fragments are associated with angiogenesis and cell migration,adhesion,invasion,apoptosis and other processes.The domain of TSP-1 can interact with a variety of cell surface receptor molecules and thus has multiple functions.TSP-1 inhibits tumor angiogenesis by inhibiting the binding of growth factors to receptors or binding to a variety of adhesion molecules on the surface of vascular endothelial cells,thereby controlling the occurrence and development of tumors.Studies have also shown that the polypeptide fragments derived from TSP-1 are with anti-angiogenesis and anti-tumor functions.In this paper,we reviewed the structure,receptor and anti-angiogenesis function of TSP-1 and hope which can provide reference for the study of angiogenesis inhibitor based on TSP-1.
