IFN-λ联合化疗药物抑制食管癌T.Tn细胞增殖的实验研究

【摘要】 [目的]探讨干扰素-λ(IFN-λ)联合化疗药物对人食管癌细胞(T.Tn细胞)增殖的影响,并探索其发挥作用的可能机制。[方法]应用MTT技术检测经IFN-λ处理后T.Tn细胞的增殖情况;根据干预措施5-氟尿嘧啶(5-fluorouracil, 5-FU)、顺铂(cis-platinum, CDDP)的不同,分为未添加任何药物的T.Tn细胞组(对照组)、IFN-λ组、化疗药物组(T.Tn细胞+5-FU组或T.Tn细胞+CDDP组)、联用组(T.Tn细胞+IFN-λ+5-FU组或T.Tn细胞+IFN-λ+CDDP组)。应用四甲基偶氮唑蓝(MTT)法检测各组中T.Tn的细胞增殖活性,从而判断两者是否存在协同作用;应用Western blotting检测细胞系中Caspase-3、Caspase-8表达情况,探索IFN-λ可能的抗T.Tn细胞增殖的机制。[结果]IFN-λ能显著抑制T.Tn细胞增殖,IFN-λ联合化疗药物组的细胞增殖活性明显降低(P<0.05)。Western blotting结果显示,IFN-λ可诱导剪切形式的Caspase3、Caspase8蛋白增加。[结论]IFN-λ联用化疗药物(5-FU、CDDP)可协同抑制T.Tn细胞增殖,其作用机制可能与Caspase3、Caspase8蛋白相关。

【Abstract】 [Objective]To investigate the effect of interferon-λ(IFN-λ)combined with chemotherapy on the proliferation of human esophageal carcinoma T.Tn cells, and to explore the possible mechanism.[Methods]To detect human esophageal carcinoma T.Tn cells proliferation situation with IFN-λ by MTT technique.All cells were divided into the control group(no drug treatment),IFN-λ group, the chemotherapy drug group and combination group(IFN-λ + chemotherapy drugs),according to the different intervention measures 5-fluorouracil(5-FU),cis-platinum(CDDP).Application of methyl thiazolyl tetrazolium(MTT)was used to detect the esophageal T.Tn cell proliferation activity in the four groups and thus to determine whether there was synergy.The expression of Caspase-3 and Caspase-8 in cell lines were detected by Western blotting, and the anti-proliferation mechanism of IFN-λ was explored.[Results]IFN-λ significantly inhibited human esophageal carcinoma T.Tn cells.The proliferative activity of IFN-λ group was significantly decreased(P< 0.05).The results of Western blotting showed that IFN-λ induced an increase in clipping form of Caspase-3 and Caspase-8 proteins.[Conclusion]IFN-λ combined with chemotherapy drug(5-FU,CDDP)could inhibit the proliferation of T.Tn cells in human esophageal carcinoma.The mechanism of action might be related to Caspase-3 and Caspase-8 proteins.
