首发精神分裂症患者PRODH及HTR2A基因表达及其多态性与认知功能的相关分析

【摘要】 目的探讨脯氨酸氧化酶和5-羟色胺2A受体基因在首发精神分裂症患者血液中的表达及其多态性与认知功能的相关性。方法将92例首发精神分裂症患者设为研究组,同期健康志愿者95名设为对照组。检测两组血液中脯氨酸氧化酶和5-羟色胺2A受体表达水平,采用MATRICS共识认知成套测验评定患者的认知功能。结果研究组血液中脯氨酸氧化酶和5-羟色胺2A受体蛋白阳性表达显著高于对照组。研究组连线测验、符号编码、视觉记忆、言语记忆、语义流畅性评分显著低于对照组(P<0.01)。脯氨酸氧化酶的rs372055和rs385440基因型比较差异有统计学意义(P<0.05)。脯氨酸氧化酶的rs383964位点不同基因型患者言语记忆差异显著(P<0.05),rs450046位点不同基因型患者符号编码评分比较差异显著(P<0.05);5-羟色胺2A受体的rs6313位点不同基因型患者符号编码评分比较差异显著(P<0.05)。结论首发精神分裂症患者血液中脯氨酸氧化酶和5-羟色胺2A受体的表达明显升高,且其认知功能损害与脯氨酸氧化酶的rs450046、rs383964和5-羟色胺2A受体的rs6311密切相关,可作为精神分裂症患者认知功能预后评估的重要分子生物学指标。

【Abstract】 Objective To investigate the expression of proline oxidase(PRODH) and 5-Hydroxytryptamine receptor 2A(HTR2A)in first-episode schizophrenia and the correlation between their polymorphisms and cognitive function.Methods A total of 92 patients with first episode schizophrenia were set as the study group,and 95 healthy volunteers during the same period were set as the control group.The expression levels of PRODH and HTR2A in the blood of patients in both groups were detected,and the cognitive function of the patients was evaluated by the MATRICS consensus cognitive battery(MCCB).Results The positive expression of PRODH and HTR2A protein in the blood of the study group was significantly higher than that of the control group.The scores of trail making test(TMT),symbol coding(SC),brief visual memory test(BVMT),hopkins verbal learning test-revised(HVLT-R),and category fluency(CF)of the study group were significantly lower than those of the control group(P<0.01).There was statistically significant difference between rs372055 and rs385440 genotypes of PRODH(P<0.05).There were significant differences in speech memory among patients with different genotypes of PRODH rs383964(P<0.05),and significant differences in symbol coding among patients with different genotypes of rs450046(P<0.05).There were significant differences in the symbol coding of different genotypes of rs6313 in HTR2A(P<0.05).Conclusions The expression of PRODH and HTR2A in blood of first-episode schizophrenia is significantly increased,and the cognitive impairment is closely re-lated to rs450046,rs383964 of PRODH and rs6311 of HTR2A,which can be used as an important molecular biological indicator for the prognosis evaluation of cognitive function in patients with schizophrenia.
