驯化对玉米Rubisco活化酶基因RCAβ序列变异的影响

【摘要】 对20份大刍草和80份玉米自交系RCAβ基因的启动子区和编码区进行测序,进行核苷酸序列变异、氨基酸序列变异、DNA多样性以及最小生成树分析,并对49份大刍草和201份玉米RCAβ基因启动子区的2个与基因表达量显著相关的位点进行单倍型分析。结果表明:在大刍草向玉米的驯化过程中,RCAβ基因受到强烈选择,导致其DNA序列变异程度降低。与玉米相比,大刍草RCAβ基因的启动子类型和编码蛋白质种类更加丰富。在玉米中未检测到T02、T03、T04和T05等4种功能域完整的大刍草RCAβ蛋白和Hap4类型大刍草启动子。大刍草RCAβ基因丰富的序列变异可为改良玉米光合效率提供遗传资源。

【Abstract】 The promoter and coding sequences of RCAβ gene in 20 teosinte individuals and 80 maize inbred lines were sequenced and underwent analysis of nucleotide variation, amino acid variation, DNA diversity and minimum spanning tree. We performed haplotype analysis on two sites in the promoter region of RCAβ gene that were significantly related to gene expression in 49 teosinte individuals and 201 maize inbred lines. The results showed that the RCAβ gene was strongly selected during the domestication from teosinte to maize, which resulted in the reduced variation of its DNA sequence. Compared with maize, the types of promoter and encoding protein of RCAβ gene in teosinte were more abundant. Teosinte RCAβ proteins(T02, T03, T04 and T05) with complete functional domains and Hap4 teosinte promoter haplo type were not detected in maize. The abundant sequence variation of RCAβ gene in teosinte could provide genetic resources for improving maize photosynthetic efficiency.
