消化时间对鱼肉中囊蚴分离提纯效果的影响研究

【摘要】 为筛选适合淡水鱼体内囊蚴分离纯化的最佳条件,从齐齐哈尔市场采集淡水鱼,压片法初筛出阳性鱼,分离鱼肉混匀称重分4份,每份取25 g,置于37℃温箱中,用人工消化液分别消化4、8、12、16 h后,离心沉淀,随机吸取50μL沉渣镜检,观察囊蚴数量及其活性,确定分离纯化效果。结果发现,消化4 h的沉渣液体浑浊,囊蚴不易分辨;3种不同类型的吸虫囊蚴消化8 h组获得囊蚴数目最多且所获囊蚴活性较好。

【Abstract】 To investigate the optimal condition for separation and purification of metacercaria from freshwater fish, freshwater fish were sampled from Qiqihaer vegetable market, and initially screened the positive fish by slide printing. The fish meat samples were digested in vitro at 37℃ digestion solutions for 4, 8, 12 and 16 hours respectively. After centrifugation, 50 μL sediments were submitted to examine metacercaria activity and counts under microscopes. After 4 hours digestion, the mixed solutions were shown too turbid to distinct metacercaria, while 8-hours digestion can result in distinctive metacercaria with high activity. Therefore, the optimal digest time for metacercaria isolation from freshwater fish might be 8 hours.
