PCV2感染仔猪的肠道黏膜免疫观察

【摘要】 猪圆环病毒(porcine circovirus,PCV)是全世界猪感染率最高的病毒,也是猪最易感染的病毒,可以感染各个阶段的猪群和猪的多数器官。其中猪圆环病毒2型(PCV2)是该病毒致病的主要血清型,是导致仔猪断奶后多系统衰竭综合征和圆环病毒相关疾病的主要病原。为进一步研究PCV2感染后宿主免疫反应特点和规律,本试验对来源于40~50日龄的22份保育猪PCV2阳性样品进行免疫学分析和流行病学回顾调查。结果表明,14份样品采样仔猪有明显的腹泻、发热或呼吸道症状等临床表现(症状明显组);8份样品采样仔猪仅有轻微腹泻或发热表征(轻微症状组)。肠道免疫组化显示,所有22份样品对应仔猪肠道中均没有检测出IgM和IgG抗体,但IgA抗体在轻微症状组的表达显著高于症状明显组。提示增强肠道粘膜免疫有利于减轻PCV2感染后带来的腹泻和肠道炎症疾病。

【Abstract】 Porcine circovirus(PCV) is the most susceptible virus to pigs, with the highest infection rate in pigs worldwide and can infect pigs at all stages and almost all organs of pigs. Porcine circovirus type 2(PCV2) is the main serotype of the virus as well as the main pathogen of post-weaning multisystem failure syndrome and circovirus-related diseases. In order to further study the characteristics and laws of host immune response after PCV2 infection, this experiment investigated the immune response and epidemiological retrospective features on 22 PCV2 positive samples isolated from nursery pigs at 40-50 days of age. The results showed that 14 piglets had obvious clinical manifestations such as diarrhea,fever or respiratory symptoms(significant symptom group); 8 piglets only displayed mild diarrhea or fever(mild symptom group). Intestinal immunohistochemistry showed that no IgM and IgG antibodies were detected in the intestines of the corresponding piglets in all samples from 22 PCV2 positive piglets, but the expression of IgA antibodies in the mild symptom group was significantly higher than that in the obvious symptom group. The results suggest that the induction of local intestinal mucosal immunity and IgA production is beneficial to alleviate diarrhea and intestinal inflammatory diseases caused by PCV2 infection.
