版纳微型猪近交系ZPBP2基因的克隆及生物信息学分析

【摘要】 为了对版纳微型猪近交系(Banna Minipig Inbred Line, BMI)ZPBP2基因进行克隆及相关生物信息学分析,该研究以BMI睾丸组织为试验材料提取RNA,通过RT-PCR方法扩增获得ZPBP2基因编码区序列,采用在线分析软件进行生物信息学分析。结果表明,ZPBP2基因编码蛋白分子量为17.4798 kD,理论等电点为5.63;在氨基酸组成上,碱性氨基酸(Lys+Arg)有14个,酸性氨基酸(Asp+Glu)有19个。在核苷酸相似度上与普通猪相似度最高,与人相似度较低;构建ZPBP2基因系统进化树的结果表明,与BMI遗传距离最近的是非近交系猪,最远是绵羊。该研究结果可为进一步全面分析和研究BMI ZPBP2基因功能及其应用奠定基础。

【Abstract】 To clone the ZPBP2 gene of Banna Mini-pig Inbred Line(BMI) and analyze its related bioinformatics, in this study, RNA was extracted from boar testis of BMI, the coding region sequences of ZPBP2 gene was amplified by RT-PCR, and the bioinformatics was analyzed by online analysis software. The results showed that the protein molecular weight of amino acid sequence was 17.4798 kD and theoretical isoelectric point was 5.63. Regarding the composition of amino acid, there were 14 alkaline amino acids(Lys+Arg) and 19 acidic amino acids(Asp and Glu).The similarity of nucleotide between BMI and common pig was highest, but the similarity between BMI and human was low. The results of ZPBP2 gene molecular phylogenetic tree showed that the closest genetic distance was BMI and common pig, the furthest was sheep. This research may establish a foundation for further study of the function and its application of ZPBP2 gene in BMI.
