质谱法检测类风湿性关节炎患者血清单糖类生物标志物
Detection of monosaccharide biomarkers in the serum of rheumatoid arthritis patients by UHPLC-MS/MS
【摘要】 目的检测类风湿关节炎(rheumatoid arthritis, RA)患者血清中4种糖类(木糖、鼠李糖、果糖和葡萄糖)生物标志物的含量。方法采用超高效液相色谱-串联质谱(ultra-high performance liquid chromatography-mass spectrometry/mass spectrometry, UHPLC-MS/MS)电喷雾离子源的多反应监测(multiple reaction monitoring, MRM)模式,分析血清样本中4种糖的含量。通过受试者工作特征(receiver operating characteristics, ROC)曲线分析木糖、鼠李糖、果糖和葡萄糖的诊断效果。结果 75例RA患者血清中木糖、鼠李糖、果糖、葡萄糖平均含量分别为12.07μg/mL、7.89μg/mL、16.89μg/mL、316.83μg/mL;50例健康人群血清中4种单糖平均含量分别为9.03μg/mL、1.83μg/mL、7.83μg/mL、149.78μg/mL。4种单糖的曲线下面积(AUC)值分别为0.635、0.671、0.567、0.704。结论 RA患者血清中4种单糖的含量均高于健康人群,其中葡萄糖AUCROC值最大,诊断价值最佳。
【Abstract】 Objective To determine the contents of four monosaccharides(xylose, rhamnose, fructose and glucose) in the serum of rheumatoid arthritis(RA)patients. Methods The ultra-high performance liquid chromatography-mass spectrometry/mass spectrometry(UHPLC-MS/MS) together with ESI source and multiple reaction monitoring(MRM) technique were used to analyse the content of total xylose, rhamnose, fructose and glucose in serum. The receiver operating characteristice(ROC) curve was used to analyse the diagnostic effect of xylose, rhamnose, fructose and glucose. Results The content of xylose, rhamnose, fructose and glucose in the serum of 75 patients were 12.07 μg/mL, 7.89 μg/mL, 16.89 μg/mL, and 316.83 μg/mL, respectively. The contents of four monosaccharides in the serum of health subjects were 9.03 μg/mL, 1.83 μg/mL, 7.83 μg/mL, and 149.78 μg/mL, respectively. The AUC values of four monosaccharides were 0.635, 0.671, 0.567, 0.704, respectively. Conclusion The contents of xylose, rhamnose, fructose and glucose in serum of RA patients are higher than those in healthy subjects. The value of glucose’s AUCROC is greatest, which has the best diagnostic value.
