白屈菜提取物的抗肿瘤作用与免疫调节作用的机制研究

【摘要】 目的研究白屈菜提取物血根碱对小鼠胃癌细胞的抑制作用及对小鼠免疫力的影响。方法通过MTT实验、流式实验、细胞迁移实验和细胞侵袭实验研究不同浓度的血根碱对小鼠胃癌细胞增殖、凋亡、迁移及侵袭的影响;通过皮下注射小鼠胃癌细胞建立BALB/c荷瘤小鼠模型,观察给予不同浓度血根碱灌胃给药30 d后对荷瘤小鼠体重、肿瘤体积、肿瘤重量及对小鼠免疫力的影响。结果血根碱(200、400μg·mL-1)明显抑制了小鼠胃癌细胞增殖(P<0.001),诱导胃癌细胞的凋亡(P<0.05),显著性抑制胃癌细胞的迁移、侵袭(P<0.01);在动物水平上与对照组比较,阳性药5-FU(10 mg·kg-1)和血根碱(200、600 mg·kg-1)均有效抑制了荷瘤小鼠肿瘤的生长(P<0.001、P<0.01);小鼠白细胞数和脾淋巴细胞转化实验结果显示,与对照组相比5-FU组(10 mg·kg-1)小鼠白细胞数和脾淋巴转化能力显著下降(P<0.01或P<0.001),血根碱组(70 mg·kg-1)小鼠白细胞数和脾淋巴转化能力显著升高(P<0.001)。结论血根碱具有免疫刺激和肿瘤抑制作用。

【Abstract】 Objective To study the inhibitory effect of Chelidonium majus extract, sanguinarine on mouse gastric cancer cells and its effects on the immunity of mice. Methods Mouse gastric cancer cells in routine cultures were treated with different concentrations of sanguinarine, and the changes in proliferation, apoptosis, cell migration and invasion of gastric cancer cells were studied by MTT assay, Muse Annexin V Dead cell kit, cell migration assay and cell invasion assay, respectively. In vivo BALB/c tumor-bearing mice were established by subcutaneous injection of cancer cells and the influence on the body weight, tumor volume, tumor weight and mice’s immunity after 30 days of intragastric administration was observed. Results Sanguinarine(200 and 400 μg·mL-1) inhibited the proliferation of mouse gastric cancer cells significantly(P<0.001), induced the apoptosis of gastric cancer cells(P<0.05) and obviously suppressed the migration and invasionof gastric cancer cells(P<0.01). The positive drug 5-FU(10 mg·kg-1) and sanguinarine(200, 600 mg·kg-1) effectively inhibited the tumor growth(P<0.001; P<0.01); Leukocyte count experiment and splenic lymphocyte transformation experiment showed that compared with control group, 5-FU(10 mg·kg-1) decreased leukocyte count and the transformation ability of mice splenic lymphocytes(P<0.01 or P<0.001), sanguinarine(70 mg·kg-1) had higher leukocyte count and transformation ability of mice splenic lymphocytes significantly compared with control group(P<0.001). Conclusion Sanguinarine has immunostimulatory and tumor suppressive effects.
