温郁金组织培养高效再生及毛状根诱导体系初探

【摘要】 温郁金为浙江省道地药材"浙八味"之一,含有抗癌药用成分β-榄香烯等.从温郁金块根上端0.1—0.2 cm处剪取长1—1.5 cm的芽尖作为外植体,利用0.1%升汞消毒10 min,在丛生芽培养基MS+6-BA 2 mg/L+IAA 1 mg/L+ZT 1 mg/L+Cef 500 mg/L上能高效获得丛生芽,芽的再生率可达100%.丛生芽经切割后置于生根培养基MS+6-BA 0.5 mg/L+IAA 0.5 mg/L上形成带根的完整植株.此外,利用发根农杆菌K599侵染温郁金组培苗的茎段、块根和根茎段,均成功诱导出毛状根;其中,根茎段毛状根的诱导率可达75%.研究结果为快速大量繁殖和培育脱毒的温郁金以及利用毛状根工厂化生长温郁金药用成分提供了基础.

【Abstract】 Curcuma wenyujin, one of the "Zhe-Ba-Wei" medicinal herbs, contains anti-cancer medicinal ingredients such as β-elemene. The tip of bud in 1—1.5 cm length is cut from 0.1—0.2 cm upper the tuberous root as the explants. After disinfection with 0.1% HgCl2 for 10 min, the tipis cultured on the medium of MS+6-BA 2 mg/L+IAA 1 mg/L+ZT 1 mg/L+Cef 500 mg/L. Clumps of buds can be efficiently differentiated and the regeneration rate of buds reach 100%. Then, the separated bud is cultured on the medium of MS+6-BA 0.5 mg/L+ IAA 0.5 mg/L, and the complete plant with roots is regenerated.Also, the hairy roots are successfully induced by Agrobacterium rhizogenes strain K599 infecting on stem segment, tuberous root and segment of root with stem. The induction rate of hairy roots from segment of root with stem can reach 75%. The results provide a basis for the rapid breeding, detoxification virus and medicinal ingredients production of Curcuma wenyujin by hairy roots.
