汉黄芩素对糖尿病肾病大鼠的血糖及肾组织TLR4、NF-κBp65水平的影响

【摘要】 目的:探究汉黄芩素对糖尿病肾病大鼠的血糖及肾组织TLR4、NF-κBp65的影响。方法:90只大鼠按照随机数字表法随机分为对照组,糖尿病肾病组,二甲双胍组及汉黄芩素低、中、高剂量组,每组15只。除对照组外,其余大鼠采用高脂饮食联合腹腔注射的方法建立糖尿病肾病大鼠模型,汉黄芩素低、中、高剂量组大鼠分别灌胃给予50、100、200 mg/kg汉黄芩素,二甲双胍组大鼠灌胃给予200 mg/kg二甲双胍,对照组及糖尿病肾病组大鼠灌胃等体积蒸馏水,1次/d,连续12周,收集并测定大鼠24 h尿量,ELISA法测定尿蛋白量、糖化血红蛋白及肾组织SOD活性、MDA水平,自动生化分析仪检测大鼠血清BUN、SCr水平,HE染色法检查大鼠肾组织病理学变化,PCR检测大鼠肾组织TLR4 mRNA、NF-κBp65 mRNA水平,Western blotting检测大鼠肾组织TLR4、NF-κBp65蛋白表达水平。结果:与糖尿病肾病组比较,汉黄芩素低、中、高剂量组大鼠24 h尿量及肾组织SOD活性明显升高(P<0.05),24 h尿蛋白量、空腹血糖、糖化血红蛋白水平、血清BUN水平、血清SCr水平、肾组织MDA水平、肾组织TLR4 mRNA水平、肾组织NF-κBp65 mRNA水平、肾组织TLR4蛋白水平及肾组织NF-κBp65蛋白水平均明显降低(P<0.05),大鼠肾组织病理学明显改善。结论:汉黄芩素能够显著改善糖尿病肾病大鼠肾功能及血糖水平,减轻大鼠肾组织氧化应激损伤及病理学变化,其机制可能与调节TLR/NF-κB信号通路有关。

【Abstract】 Objective: To study the effects of wogonin on blood glucose and renal tissue TLR4, NF-κB p65 in rats with diabetic nephropathy. Methods: According to the random number table method, 90 rats were randomly divided into control group, diabetic nephropathy group, metformin group, wogonin low, medium and highdose group, with 15 rats in each group. Except for the control group, the diabetic nephropathy rat model was established by high fat diet combined with intraperitoneal injection. The wogonin low, medium and high-dose group were given 50, 100 and 200 mg/kg wogonin respectively. The rats in the metformin group were given metformin according to 200 mg/kg, and the rats in the control group and diabetic nephropathy group were given normal saline once a day for 12 weeks. The 24-hour urine volume, fasting blood glucose and glycosylated hemoglobin levels, serum urea nitrogen(BUN) and serum creatinine(SCr) levels, renal superoxide dismutase(SOD) activity and malondialdehyde(MDA) level were measured. The pathological changes of renal tissue were detected by hematoxylin-eosin(HE) staining. The levels of TLR4 mRNA and NF-κB p65 mRNA in renal tissue were detected by real-time quantitative polymerase chain reaction(PCR), and the levels of TLR4 and NF-κB p65 in renal tissue were detected by Western blotting. Results: Compared with diabetic nephropathy group, 24-hour urine volume and renal SOD activity were significantly increased(P<0.05), 24-hour urinary protein, fasting blood glucose and glycosylated hemoglobin levels, serum BUN and SCr levels, renal tissue MDA levels, renal tissue TLR4 m RNA, NF-κB p65 m RNA levels and TLR4, NF-κB p65 protein levels were significantly decreased in wogonin groups(P<0.05). Renal histopathology was significantly improved. Conclusion: Wogonin can significantly improve renal function and blood glucose levels in diabetic nephropathy rats, and reduce oxidative stress damage and pathological changes in rat renal tissue. The mechanism may be related to the regulation of TLR/NF-κB signaling pathway.
