腱糖蛋白C在脑胶质瘤中的表达及临床意义

【摘要】 目的分析腱糖蛋白C(TNC)在脑胶质瘤中的表达及临床意义。方法回顾性分析中国脑胶质瘤基因组图谱(CGGA)数据库中胶质瘤患者转录组数据和临床资料,采用SPSS 22.0统计软件分析TNC在胶质瘤组织中的表达特点,分析TNC表达水平与患者临床特征及预后的关系。结果 TNC在胶质瘤组织中表达水平高于正常对照脑组织(P<0.05);TNC表达水平在胶质瘤组织中随世界卫生组织(WHO)病理分级增高而升高(P<0.05)。TNC在异柠檬酸脱氢酶(IDH)基因野生型、年龄>42岁、O~6-甲基鸟嘌呤DNA甲基转移酶(MGMT)基因启动子非甲基化患者的脑胶质瘤组织中高表达(P<0.05)。TNC高表达组患者总生存期均短于TNC低表达组患者(P<0.05)。多因素Cox回归分析结果显示,病理分级、术后化疗、1p/19q染色体共缺失和TNC表达水平是脑胶质瘤患者生存期的独立影响因素(P<0.05)。结论 TNC在脑胶质瘤中的表达水平与脑胶质瘤的恶性程度相关,可作为脑胶质瘤的诊断标志物及预后预测因子。

【Abstract】 Objective To analyze the expression and clinical significance of tenascin C(TNC) in glioma.Methods The clinical and sequencing data of patients with glioma from the Chinese Glioma Genome Atlas(CGGA) database were retrospectively analyzed. The SPSS 22.0 statistical software was used to analyze the expression characteristics of TNC in glioma tissue, and the relationships between the expression level of TNC and clinical characteristics and prognosis of patients were analyzed.Results The expression level of TNC in glioma tissue was higher than that in normal control brain tissue(P<0.05), and the expression level of TNC in glioma tissue increased with the increase of World Health Organization(WHO) pathologic grading(P<0.05). TNC was highly expressed in glioma tissues of patients with wild-type isocitrate dehydrogenase(IDH) gene, over 42 years old and unmethylated promoter of O~6-methylguanine-DNA methyltransferase(MGMT) gene(P<0.05). The overall survival time of every TNC high expression group was shorter than that in corresponding TNC low expression group(P<0.05). Multivariate Cox regression analysis showed that pathologic grading, postoperative chemotherapy, 1 p/19 q chromosome co-deletion and expression level of TNC were independent factors affecting the survival time of patients with glioma(P<0.05).Conclusion The expression level of TNC is positively correlated with the malignant degree of glioma, which can be used as a diagnostic marker and prognostic predictor of glioma.
