miR-34a-5p下调Notch1表达增强三阴性乳腺癌MDA-MB-231细胞对紫杉醇的敏感性

【摘要】 目的:本研究旨在阐明miR-34a-5p是否通过下调Notch1表达,进而增强三阴性乳腺癌MDA-MB-231细胞对紫杉醇的化疗敏感性。方法:qRT-PCR测定MCF-10A细胞与MDA-MB-231细胞miR-34a-5p的表达水平;以miR-34a mimic瞬时转染MDA-MB-231细胞,qRT-PCR与western blot分析Notch1表达情况,最后CCK-8试剂盒测定紫杉醇对于不同Notch1表达水平MDA-MB-231细胞的细胞毒作用。结果:miR-34a-5p在正常乳腺MCF-10A细胞高表达,而在三阴性乳腺癌MDA-MB-231细胞低表达;转染miR-34a mimic使MDA-MB-231细胞中Notch1表达降低,敲低MDA-MB-231细胞Notch1表达降低紫杉醇抑制细胞存活力作用的IC50。结论:miR-34a-5p通过下调Notch1表达,进而增强紫杉醇抑制三阴性乳腺癌MDA-MB-23 1细胞存活力作用。

【Abstract】 Objective This study aims to clarify whether miR-34a-5p downregulated the expression of Notch1 and enhanced the sensitivity of triple negative breast cancer MDA-MB-231 to Paclitaxel.Methods The expression of miR-34a-5p in MCF-10A and MDA-MB-231 was determined by Quantitative Real-time PCR (qRT-PCR);MDA-MB-231 were transfected with miR-34a mimic and the expression of Notch1was analyzed by qRT-PCR and western blot;IC50 of Paclitaxel for MDA-MB-231 that different expression status of Notch1 was determined by CCK-8 assay.Results miR-34a-5p is highly expressed in normal breast cell MCF-10A than in triple negative breast cancer MDA-MB-231cell line;the expression of Notch1 was downregulated by transfected with miR-34a mimic in MDA-MB-231 cells;and IC50 of Paclitaxel was significantly lower in MDA-MB-231 cells in which Notch1 expression was knocked down in contrast to the control group.Conclusion Sensitivity of a triple negative breast cancer MDA-MB-231 cell line to paclitaxel was enhanced by down-regulating Notch1 expression through upregulating miR-34a-5p.
