H1foo在哺乳动物卵子发生和早期胚胎发育中的表达及功能研究进展
Review on the expression pattern and function of H1foo during mammalian oogenesis and early embryonic development
【摘要】 H1连接组蛋白(H1 linker histone,H1)是染色质结构和功能的关键调节因子。在相邻的核小体之间,H1与连接体DNA(linker DNA)结合构成染色质的念珠状结构,以稳定染色质的高级结构、调节基因表达。研究发现一种H1亚型,命名为卵母细胞特异性连接组蛋白(oocytespecific linker histone,H1foo),在哺乳动物卵母细胞中特异性表达。文章主要针对H1foo在卵巢卵泡、卵母细胞及早期胚胎发育中的表达分布模式,H1foo在卵母细胞减数分裂恢复和早期胚胎发育中的作用,H1foo在卵母细胞受精和体细胞核移植过程中与其他成分置换、实现基因组的完全重编程、恢复全能性中的重要作用进行综述,以期为相关机制研究提供参考。
【Abstract】 Linker histones H1s are key regulators of chromatin structure and function.The linker histones,in turn,constitute the major proteins bound,in part,to linker DNA,the DNA-bridging nucleosome core particles.The rosary-like structure of chromatin is formed.Linker histones play a crucial role in the higher-order packaging of chromatin and regulating of gene expression.A subtype of H1,named oocyte-specific linker histone H1foo,was found to be specifically expressed in mammalian oocytes.This study focuses on the expression and distribution pattern of H1foo in ovarian follicles,oocytes andearlyembry.Theroleof H1fooinoocytemeiosisrecoveryandearlyembryonicdevelopment,thereplacementofprotamine and somatic H1 with H1foo in the process of fertilization and somatic cell nuclear transfer,the reprogram of nuclear genome and the restoration of totipotency are reviewed,in order to provide reference for the relevant mechanism research.
