MAL2调控SGK1通路活性促进肺癌细胞的增殖和转移
MAL2 promoting the proliferation and metastasis of lung cancer cells by regulating the activity of the SGK1 pathway
【摘要】 目的探讨MAL2(myelin and lymphocyte protein 2)在肺癌中的功能及作用机制。方法分析TCGA数据中MAL2在肺癌组织中的表达情况,在H1299和A549细胞中用克隆形成实验检测MAL2基因敲减后对肺癌细胞增殖的影响,探讨MAL2在肺腺癌细胞增殖中的作用,并用CCK8等细胞增殖、转移、凋亡实验进一步验证其功能;将MAL2干扰后,采用Western blot实验检测下游通路基因表达情况以分析MAL2基因可能的作用机制。结果 MAL2基因在肺癌组织与癌旁组织相比表达显著升高(P<0.05),且MAL2敲减后抑制细胞增殖和转移、促进细胞凋亡(P<0.05);MAL2可能通过调控SGK1通路发挥作用。结论 MAL2基因在肺癌的恶性进展中起重要的作用,该作用通过调控SGK1机制进行,MAL2基因是肺癌的一个新的潜在的分子诊断指标及治疗靶点。
【Abstract】 Objective To investigate the function and mechanism of myelin and lymphocyte protein 2(MAL2) in lung cancer.Methods The expression of MAL2 in lung cancer tissues in TCGA data was analyzed, and clone formation experiments in H1299 and A549 cells were conducted to detect the effect of MAL2 gene knockdown on the proliferation of lung cancer cells, so as to explore the role of MAL2 in the proliferation of lung adenocarcinoma cells. CCK8 assay for cell proliferation, metastasis, and apoptosis was performed to further verify its function.After MAL2 interference, Western blot experiment was used to detect the expression of downstream pathway genes, so as to analyze the possible mechanism of MAL2 gene.Results The expression of MAL2 gene in lung cancer tissues was significantly higher than that in adjacent tissues(P<0.05), and knockdown of MAL2 inhibited cell proliferation and metastasis, and promoted cell apoptosis(P<0.05); MAL2 might play a role by regulating the SGK1 pathway.Conclusion The MAL2 gene plays an important role in the malignant progression of lung cancer by regulating the SGK1 mechanism. Therefore, MAL2 gene is a novel and potential molecular diagnostic index and therapeutic target for lung cancer.
