circ_0084927靶向miR-623调控喉癌TU177细胞增殖和凋亡的机制研究

【摘要】 目的探讨circ0084927靶向miR-623对喉癌TU177细胞增殖和凋亡的影响。方法选取2017年6月—2019年6月接受手术治疗的喉癌41例,检测癌组织和癌旁组织circ0084927和miR-623的表达水平。将喉癌TU177细胞分为si-NC组、si-circ0084927组、miR-NC组、miR-623组、pcDNA组、pcDNA-circ0084927组、si-circ0084927+anti-miR-NC组和si-circ0084927+anti-miR-623组,检测细胞光密度(OD)值、凋亡率、细胞克隆形成数及凋亡相关蛋白表达情况;应用双荧光素酶报告实验确定circ0084927和miR-623的靶向关系。结果与癌旁组织比较,喉癌组织中circ0084927相对表达水平升高,miR-623相对表达水平降低(P<0.05)。与si-NC组比较,si-circ0084927组TU177细胞OD值、克隆形成数降低,凋亡率及剪切型半胱氨酸蛋白酶3(Cleaved-caspase3)、Cleaved-caspase9蛋白表达水平升高(P<0.01)。与miR-NC组比较,miR-623组TU177细胞OD值、克隆形成数降低,凋亡率及Cleaved-caspase3、Cleaved-caspase9蛋白表达水平升高(P<0.01)。与pcDNA组比较,pcDNA-circ0084927组TU177细胞miR-623相对表达水平降低(P<0.05)。与si-circ0084927+anti-miR-NC组比较,si-circ0084927+anti-miR-623组TU177细胞OD值、克隆形成数升高,凋亡率及Cleaved-caspase3、Cleaved-caspase9蛋白表达水平降低(P<0.05)。结论干扰circ0084927通过靶向上调miR-623表达可抑制喉癌TU177细胞增殖,并诱导细胞凋亡。

【Abstract】 Objective To explore the effects of circ_0084927 targeting miR-623 on proliferation and apoptosis of laryngeal cancer TU177 cells by targeting miR-623. Methods A total of 41 patients with laryngeal cancer who had undergone surgeries between June 2017 and June 2019 were selected, and expressions of circ_0084927 and miR-623 in cancer tissues and adjacent tissues were detected. TU177 laryngeal cancer cells were divided into si-NC group, si-circ_0084927 group, miR-NC group, miR-623 group, pcDNA group, pcDNA-circ_0084927 group, si-circ_0084927+anti-miR-NC group and si-circ_0084927+anti-miR-623 group, and cell optical density(OD) value, apoptosis rate, cell clone formation number and the expressions of apoptosis-related proteins were detected. The targeting relationship between circ_0084927 and miR-623 was determined by double luciferase reporting experiment. Results Compared with those in adjacent tissues, the relative expression of circ_0084927 was significantly increased, while the relative expression of miR-623 was significantly decreased in laryngeal cancer tissues(P<0.05). Compared with those in si-NC group, the OD value and the colony formation numbers of TU177 cells were significantly decreased, while apoptosis rate and expressions of Cleaved-caspase3 and Cleaved-caspase9 proteins were significantly increased in si-circ_0084927 group(P<0.01). Compared with those in miR-NC group, the OD value and colony formation numbers of TU177 cells were significantly decreased, while apoptosis rate and expressions of Cleaved-caspase3 and Cleaved-caspase9 proteins were significantly increased in miR-623 group(P<0.01). The relative expression of miR-623 in TU177 cells in pcDNA-circ_0084927 group was significantly lower compared with that in pcDNA group(P<0.05). Compared with those in si-circ_0084927+anti-miR-NC group, the OD value and colony formation numbers of TU177 cells were significantly increased, while apoptosis rate and expressions of Cleaved-caspase3 and Cleaved-caspase9 proteins were significantly reduced in si-circ_0084927+anti-miR-623 group(P<0.05). Conclusion Interfering with circ_0084927 may inhibit the proliferation and induce cell apoptosis of laryngeal carcinoma TU177 cells by targeting and up-regulating miR-623.
