肾茶提取物对小鼠三聚氰胺肾结石模型的影响

【摘要】 目的探讨肾茶提取物对三聚氰胺及其同系物三聚氰酸所致肾结石的防治作用。方法 40只雄性ICR小鼠随机分为正常对照组、模型组、肾茶组和阳性对照组,以三聚氰胺和三聚氰酸混悬液造模,分别给予生理盐水、肾茶提取液、枸橼酸氢钾钠溶液,连续灌胃15 d后,检测小鼠的BUN、Cr指标,HE染色观察肾脏病理形态学改变,LC-MS/MS测定三聚氰胺在肾脏组织中的含量。结果与正常对照组比较,染毒第15 d模型组小鼠体质量增长明显减慢(P<0.05);与模型组比较,肾茶组、阳性对照组同日组间体质量增加,但统计学无显著差异(P>0.05)。染毒15 d后,小鼠血清BUN、Cr各组间统计学无显著差异。三聚氰胺在小鼠肾脏组织中的含量:模型组(92.1 ng·g-1)>阳性对照组(88.5 ng·g-1)>肾茶组(26.1 ng·g-1)>正常对照组(0 ng·g-1)。肾脏病理:与正常对照组比较,模型组小鼠肾小管腔内可见结晶形成,肾茶组肾小管内偶见少许沉积物,无结晶形成,枸橼酸氢钾钠组肾小管内可见沉积物,偶见结晶。结论肾茶提取物有抑制三聚氰胺和三聚氰酸肾结石形成的作用。

【Abstract】 OBJECTIVE To explore the possible effect of Clerodendranthus spicatus extracts on the prevention and treatment of kidney stones induced by melamine and its homologues cyanuric acid.METHODS 40 ICR male mice were randomly divided into four groups: normal control group, model group, C.spicatus group and positive control group.Kidney stones model was induced by oral gavage administration of melamine and cyanuric acid mixture at 8 a.m.In the afternoon, each group was given normal saline, Clerodendranthus spicatus extract [1 g/(kg·d)] and potassium sodium hydrogen citrate solution [2.5 g/(kg·d)] respectively.After 15 days the blood was collected for biochemical analysis and kidneys were taken for HE staining to evaluate the renal pathological and LC-MS/MS determination of melamine in the tissue of the kidney.RESULTS Compared with normal control group, the body weight growth of the model group was significantly slowed down at the 15 d(P<0.05).Compared with the model group, the C.spicatus group and positive control group increased on the same day, but there was no statistically significant difference(P>0.05).There were not significant different between each group in serum biochemical after 15 days exposure of a mixture of melamine and cyanuric acid.The measured results of melamine in the kidneys of mice: model group(92.1 ng·g-1)>positive control group(88.5 ng·g-1)>C.spicatus group(26.1 ng·g-1)>normal control group(0 ng·g-1).Compared with the normal group, crystal formation was observed in the renal tubules of mice in the model group, occasionally a little sediment was observed in the renal tubules of the C.spicatus group without crystal formation, and occasionally crystals were observed in the renal tubules of the positive control group.CONCLUSION Clerodendranthus spicatus extract play a renoprotective role in melamine and cyanuric acid-induced kidney stone in mice.
