基于特征图谱和OPLS-DA的茯苓产地溯源分析

【摘要】 目的分析不同产地茯苓的成分差异,为茯苓的产地溯源研究提供参考。方法采用Sepax HP-C18柱(4.6 mm×150 mm, 5μm),以0.5%乙酸水溶液(A)-乙腈(B)为流动相进行梯度洗脱,建立特征图谱,结合正交偏最小二乘法判别分析(OPLS-DA)筛选不同产地茯苓的差异成分。结果建立了茯苓的特征图谱,标定并归属21个共有峰和9个非共有峰;通过OPLS-DA共筛选出7种差异成分,包括3种共有成分和4种非共有成分。结论通过OPLS-DA分析茯苓特征图谱的差异,并筛选出7种差异成分可用于茯苓产地溯源研究。

【Abstract】 OBJECTIVE To analyze the compositional difference of Poria cocosand provide a reference for the origin traceability of Poria cocos.METHODS Sepax HP-C18(4.6 mm×150 mm, 5 μm) column was applied to the establishment of the fingerprint of Poria cocos, with acetonitrile-water(0.5% acetic acid) as mobile phase.Then, screen out the different components inPoria cocos from different habitatsby orthogonal partial least squares discriminant analysis(OPLS-DA).RESULTS A total of 21 common peaks and 9 non-common peaks were identified inthe fingerprint of Poria cocos.Through OPLS-DA screened out 7 different components, including 3 common components and 4 non-common components.CONCLUSION OPLS-DA was used to analyze the differences in the fingerprint, and screened out 7 different components that could be used for the traceability of Poria cocos.
