隐丹参酮抗大鼠吗啡条件性位置偏爱形成及其对相关脑区信号分子水平的影响

【摘要】 目的探究隐丹参酮对大鼠吗啡条件性位置偏爱(CPP)效应形成的影响及其抗吗啡CPP的可能作用部位和机制。方法建立吗啡CPP效应模型观察隐丹参酮对其影响;实时荧光定量PCR和Western Blot法检测大鼠成瘾相关脑区信号分子水平变化。结果:中剂量隐丹参酮40 mg·kg-1干预组可拮抗吗啡CPP效应形成;该组内侧前额叶皮层及海马区BDNF的表达水平与吗啡组比较明显降低。结论中剂量隐丹参酮40 mg·kg-1可抑制吗啡CPP效应形成;这种抑制可能与其减少BDNF信号分子表达水平有关。

【Abstract】 OBJECTIVE To investigate the effect of cryptotanshinone on the formation of morphine-induced conditioned place preference(CPP) in rats and the possible role and mechanism.MEHTODS Intraperitoneal injection of cryptotanshinone was used to observe its effect on the formation of CPP induced by morphine;Western Blot and Real-time quantitative PCR were used to detect the level of signal molecules in related brain regions.RESULTS Cryptotanshinone at a dose of 40 mg·kg-1 could inhibit the formation of morphine CPP;Compared with morphine group,the level of BDNF in the mPFC and Hippo were significantly decreased in the cryptotanshinone at a dose of 40 mg·kg-1 intervene group by western-blot and RT-PCR.CONCLUSION Cryptotanshinone at a dose of 40 mg·kg-1 showed inhibiting the formation of morphine-induced CPP in rats;The inhibition may be related to decrease the level of BDNF signaling molecules.
