上皮间质转化相关标志物在不同类型慢性鼻窦炎患者中的表达

【摘要】 目的:研究上皮间质转化(EMT)相关标志物在不同类型慢性鼻窦炎(CRS)患者中的表达。方法:收集2019年9月至2020年5月在东莞市人民医院耳鼻咽喉科行鼻内镜手术治疗的CRS患者组织40例,根据内型分为非嗜酸性粒细胞型CRS(Non-ECRS)及嗜酸性粒细胞型CRS(ECRS),每组20例。同期选取无CRS行鼻内镜手术患者15例为正常对照组。免疫组织化学染色和Western blotting法检测各组上皮细胞表型标志E-钙黏蛋白(E-cadherin)、闭合蛋白(Occludin)和封闭蛋白4(Claudin-4)以及间质细胞表型标志物α-平滑肌肌动蛋白(α-SMA)、N-钙粘附蛋白(N-cadherin)、波形蛋白(Vimentin)的表达。结果:与对照组相比,Non-ECRS组和ECRS组鼻黏膜上皮细胞中E-cadherin、Occludin和Claudin-4表达均明显下调,α-SMA、vimentin和N-cadherin表达均明显上调(均P<0.05),ECRS组中α-SMA、Vimentin和N-cadherin表达低于Non-ECRS组(P<0.05)。结论:不同类型的CRS黏膜上皮细胞中均存在EMT现象,以Non-ECRS鼻黏膜中更为明显。EMT可能与CRS发病机制有关。

【Abstract】 Objective: To investigate the epithelial-mesenchymal transition(EMT) related biomarkers in the subtypes of chronic rhinosinusitis(CRS). Methods: A total of 40 patients with chronic rhinosinusitis in our hospitalfrom September 2019 to May 2020 were selected, and divided into non-eosinophilic chronic rhinosinusitis(ECRS)group and ECRS group. Fifteen patients without CRS underwent nasal endoscopic surgery were defined asacontrol group.Immunohistochemisty staining and Western blotting were applied to detect the expressions of epithelial markers such as E-cadherin, Occludinand Claudin-4 and mesenchymal markers such as α-SMA,VimentinandN-cadherin. Results: The expressions of E-cadherin, Occludin and Claudin-4 were significantly down-regulated in non-ECRSand ECRS groups, while the expressions of α-SMA,Vimentin and N-cadherin were significantly up-regulated compared with the control group(P<0.05). Furthermore, the expressions of α-SMA,Vimentin and N-cadherin were lower in ECRS group than those innon-ECRS group(P<0.05). Conclusion: EMT exists in different types of CRS mucosal epithelial cells, particularly in non-ECRS, indicating that EMT may be related to the pathogenesis of CRS.
