TREM1/TREM2在小鼠呼吸机相关性肺损伤中的表达
The expression of TREM-1/TREM-2 in ventilator-induced lung injury in mice
【摘要】 目的:探讨髓样细胞触发受体(TREM)1和TREM2在小鼠呼吸机相关性肺损伤(VILI)模型中的表达情况。方法:将30只C57BL/6小鼠随机分为自主呼吸组(CON组)、正常潮气量组(NVT组,VT=10 mL/kg)、大潮气量组(HVT组,VT=40 mL/kg),其中HVT组分为HVT 1[机械通气(MV)1 h]、HVT 2(MV 2 h)、HVT 3(MV 4 h)3个亚组。测定肺组织湿/干重(W/D)比值,苏木精—伊红(HE)染色观察小鼠肺组织病理改变,酶联免疫吸附试验(ELISA)法检测BALF中总蛋白、白介素(IL)-1β、IL-6的含量,分别采用实时荧光定量PCR(qPCR)法和Western blotting法检测肺组织TREM1、TREM2 mRNA和蛋白的表达。结果:与CON组比较,HVT 2组和HVT 3组肺组织W/D比值、肺组织病理学评分及BALF中总蛋白、IL1-β、IL-6水平均明显升高(均P<0.05)。与CON组相比,HVT 1组、HVT 2组和HVT 3组肺组织TREM1 mRNA和蛋白表达显著增高,TREM2 mRNA和蛋白表达显著降低,TREM1/TREM2比值显著高于CON组和NVT组(均P<0.05)。TREM1与TREM2表达呈负相关关系(P<0.05)。结论:小鼠VILI中,促炎因子TREM1与抗炎因子TREM2比值显著上调,TREM1/TREM2比值可以动态评价肺组织炎症变化。
【Abstract】 Objective: To investigate the expression of trigger receptor expressed on myeloid cells 1/2(TREM1/2) in ventilator-related lung injury(VILI) in mice. Methods: Thirty C57 BL/6 mice were randomly divided into spontaneous breathing group(CON group), normal tidal volume group(NVT group, VT=10 mL/kg), and high tidal volume group(HVT group, VT=40 mL/kg). HVT group was divided into HVT1(mechanical ventilation for1 h), HVT2(mechanical ventilation for 2 h), and HVT3(mechanical ventilation for 4 h) subgroups. The lung wet/dry weight(W/D) ratio was assessed, and the histopathology changes were observed by hematoxylin-eosin staining(HE). The total protein, the levels of interleukin(IL)-1β and IL-6 in BALF were determined by enzymelinked immunosorbent assay(ELISA). The mRNA and proteinexpression of TREM1 and TREM2 in lung tissues were measured by RT-qPCR and Western blotting, respectively. Results: Compared with the CON group, the lung W/D ratio, lung histopathological score, and the total protein, IL-1β and IL-6 levels in BALF in HVT2 and HVT3 groups were significantly higher than those in CON group(P<0.05). Compared with CON group, the mRNA and protein expression of TREM1 was significantly increased in HVT1 group, HVT2 group and HVT3 group, while the mRNA and protein expression of TREM2 was significantly decreased.The ratio of TREM1/TREM2 in HVT groups was significantly higher than that in CON group and NVT group(P<0.05). There was a negative correlation between the expression of TREM1 and TREM2(P<0.05). Conclusion:In VILI mice, the ratio of pro-inflammatory factor TREM1 and anti-inflammatory factor TREM2 was significantly up-regulated, and the ratio of TREM1/TREM2 can dynamically evaluate the changes of lung tissue inflammation.
