长寿及冠心病人群miR-27a-3p血清表达水平的状况分析
Study on the serum expression profiles of miR-27a-3p in long-lived cohort and patients with coronary heart disease
【摘要】 目的:研究miR-27a-3p在红水河流域长寿人群及广西地区冠心病人群血清中的表达状况,探讨miR-27a-3p与衰老相关性疾病的关系。方法:实时荧光定量PCR(q PCR)检测长寿组(年龄>90岁)、老年组(65~84岁)、年轻组(<65岁)、冠心病组(46~90岁)和健康对照组(50~85岁)血清中miR-27a-3p表达情况,使用Spearman评估miR-27a-3p表达水平与各临床指标的相关性。结果:miR-27a-3p在长寿组及年轻组血清中相对表达量高于老年组(P<0.05);整体分析,miR-27a-3p相对表达量与体重指数(Ibm)、空腹血糖(FBG)、收缩压(SBP)、C反应蛋白(CRP)、载脂蛋白A(Apo A)和载脂蛋白B(Apo B)呈负相关关系(P<0.05);在长寿组中,miR-27a-3p相对表达量与Ibm、SBP、舒张压(DBP)、CRP和Apo B呈负相关关系(P <0.05);在老年组中,miR-27a-3p相对表达量与DBP、CRP成负相关关系(P <0.05);在年轻组中,miR-27a-3p相对表达量与CRP、Apo B呈负相关关系(P<0.05);与正常对照组相比,miR-27a-3p在冠心病患者中低表达。结论:miR-27a-3p在红水河流域长寿人群中高表达,在冠心病样本中低表达,且miR-27a-3p表达量与多种冠心病危险因素呈负相关关系,提示该长寿人群可能因为miR-27a-3p的高表达减少了冠心病的发生而体现一定的生存优势。
【Abstract】 Objective: To study the serum expression level of miR-27 a-3 p in long-lived population from Hongshuihe River Basin and patients with coronary heart disease(CHD) from Guangxi province and evaluate the relationship between miR-27 a-3 p age-related diseases. Methods: Serum expression level of miR-27 a-3 p was detected by RT-qPCR in healthy long-lived group(>90 years), CHD group(46-90 yrs), normal elderlies(65-84 years) and youngers(<65 years) and its association with clinical parameters was evaluated by Spearman analyses. Results: The expression of miR-27 a-3 p in the serum of the longevity group and the young group was higher than that of the elderly group(P<0.05). In the pooled population, the relative expression of miR-27 a-3 p was negatively correlated with body mass index, fasting blood glucose level(FBG), systolic blood pressure(SBP), C-relative protein(CRP), apolipoprotein A(Apo A) and apolipoprotein B(Apo B)(P<0.05); the relative expression level of miR-27 a-3 p was negatively correlated with body mass index, SBP, diastolic blood pressure(DBP), CRP and Apo B in the longevity group while was negatively linked to DBP, CRP in the elderly group and to CRP and ApoB in the young group(P<0.05). MiR-27 a-3 p was lower expressed in patients with CHD as comparing with healthy controls. Conclusion: MiR-27 a-3 p is highly expressed in the serum of long-lived individuals in the Hongshuihe River Basin while lower expressed in CHD patients, which is negatively correlated with several CHD risk factors, suggesting that the long-lived population may have a survival advantage in that the high expres-sion of miR-27 a-3 p may reduce the occurrence of CHD.
