前列腺癌中HIF-1α和RBM5表达的临床研究
Clinical Significance of Hypoxia-inducible Factor-1α and RBM5 Expression in Prostate Cancer
【摘要】 目的探讨前列腺癌(PCa)中缺氧诱导因子(HIF-1α)和RBM5表达及相关性。方法采用免疫组化PV-9000两步法,半定量逆转录PCR (RT-PCR)和Western blot分别检测50例PCa患者组织标本中HIF-1α和RBM5的表达,同时检测25例正常前列腺(NP)和50例良性前列腺增生(BPH)组织标本中这两种蛋白的表达作为对照进行比较分析。结果 PV-9000两步法检测数据显示,RBM5和HIF-1α在3种不同组织中的表达差异显著,有统计学意义(P <0.01); RBM5和HIF-1α两者在同一种组织中的表达差异显著,有统计学意义(P <0.01)。RT-PCR检测结果显示,NP和BPH组织细胞中RBM5 mRNA表达阳性,而PCa中未检测到RBM5 mRNA的表达; HIF-1αmRNA在PCa组织细胞中高表达,在NP和BPH中则表达甚微。Western blot检测结果显示,在NP和BPH组织细胞中RBM5蛋白有极高表达,在PCa中只有极低表达; HIF-1α蛋白在NP和BPH组织细胞中表达水平极低,在PCa中表达水平极高。结论 HIF-1α和RBM5表达密切相关,增强RBM5表达的同时沉默HIF-1α极有可能大大提高目前分子靶向治疗肿瘤的疗效,尤其为去势抵抗PCa的治疗提供新策略。
【Abstract】 Objective To investigate the expression of hypoxia-inducible factor-1α( HIF-1α) and RBM5 in prostate cancer( PCa). Methods HIF-1α and RBM5 expression in tumor tissue of 50 patients with PCa were detected by immunohistochemical staining,reverse transcription polymerase chain reaction( RT-PCR) and Western blot. 25 normal prostate samples( NP)and 50 benign prostatic hyperplasia( BPH) samples were as control. Results Immunohistochemical staining showed that there were significant differences on the expressions of RBM5 and HIF-1α among PCa,NP and BPH( P < 0. 01),and there were significant differences on the expressions of both RBM5 and HIF-1α in the same tissue( P < 0. 01). RT-PCR showed that RBM5 mRNA expression was positive in NP and BPH,while negative in PCa; HIF-1α mRNA expression was high in PCa and slight in NP and BPH. Western blot showed that RBM5 protein level was high in NP and BPH,but very low in PCa; HIF-1α protein level in NP and BPH was extremely low,but extremely high in PCa. Conclusion HIF-1α expression might be closely related to the expression of RBM5.
