近红外光谱法快速测定白芍中3种成分的含量
Rapid Determination of Three Components in Baishao by Near-infrared Spectroscopy
【摘要】 目的:建立白芍生品中没食子酸、芍药苷及芍药内酯苷含量的快速测定近红外光谱定量模型。方法:以HPLC检测值为参照,采用傅里叶变换近红外漫反射光谱技术采集白芍生品的近红外光谱,结合偏最小二乘法(PLS)分别建立没食子酸、芍药苷及芍药内酯苷含量的快速测定方法。结果:白芍中没食子酸、芍药苷及芍药内酯苷的校正模型相关系数(R)、校正均方差(RMSEC)、内部验证均方差(RMSEP)分别为0.99132、0.0055、0.00896;0.99869、0.0266、0.10900;0.97052、0.0349、0.04210。通过外部验证,白芍的预测值与真实值相关系数分别为0.9404、0.9412和0.9446。结论:该方法准确、快速、简便,可实现大批量样品的快速分析。
【Abstract】 Objective: To establish a quantitative model for the rapid determination of gallic acid,paeoniflorin and albiflorin in raw Baishao(Radix Paeoniae Alba) by near-infrared spectroscopy. Methods: By taking the detection value of HPLC as the reference, Fourier transform near infrared diffuse reflectance spectroscopy was adopted to collect near-infrared spectroscopy of raw Baishao, the method for rapid determination of the contents of gallic acid, paeoniflorin and albiflorin was established by partial least squares(PLS). Results:Correction model correlation coefficient(R), corrected mean square error deviation(RMSEC) and mean square error deviation of internal validation(RMSEP) of gallic acid, paeoniflorin and albiflorin in Baishao were 0.99132,0.0055 and 0.00896; 0.99869, 0.0266 and 0.10900; 0.97052, 0.0349 and 0.04210 respectively. Correlation coefficients between predicted value and real value of Baishao reached 0.9404, 0.9412 and 0.9446 by external verification. Conclusion: The method, accurate, rapid and simple, could realize the rapid analysis of the samples in large scales.
