检测碱性磷酸酶荧光探针的性能研究及成像的应用

【摘要】 目的:本实验对军事医学研究院提供的检测碱性磷酸酶的荧光探针进行性能的评价及成像实验。方法:利用紫外分光光度计及荧光光谱仪测试该荧光探针的吸收光谱及发射光谱;利用CCk8试剂盒检测荧光探针对细胞的毒性;利用OLYMPUS-IX73倒置荧光显微镜测试荧光探针对碱性磷酸酶的特异性识别。结果:经紫外光谱仪及荧光光谱仪测试表明该探针具有良好的吸收峰及发射峰;较低的细胞毒性;细胞成像实验可知,能识别细胞内的碱性磷酸酶。

【Abstract】 Purpose: This experiment evaluates the performance of the fluorescent probe for detecting alkaline phosphatase provided by the Military Medical Research Institute and performs imaging experiments. Methods: The absorption and emission spectra of the fluorescent probe were measured by ultraviolet spectrophotometer and fluorescence spectrometer.CCK8 kit was used to detect the toxicity of fluorescent probe to cells. The specific recognition of alkaline phosphatase by fluorescence probe was tested by OLYMPUS-IX73 inverted fluorescence microscope.Results: Tests by ultraviolet spectrometer and fluorescence spectrometer showed that the probe has good absorption and emission peaks; low cytotoxicity; cell imaging experiments show that it can identify alkaline phosphatase in cells.
