两株低温石油烃降解菌的筛选鉴定及复配优化
Screening Identification and Compounding Optimization of Two Low Temperature Petroleum Hydrocarbon Degrading Bacteria
【摘要】 筛选能够在低温条件下降解石油烃的菌株,探究降解菌株在低温条件下对石油烃的降解能力。从克拉玛依油田长期受到石油污染的土壤中通过富集、分离和纯化得到两株低温石油烃降解菌,经过形态学、生理生化和16S rDNA初步鉴定这两株菌为Agrobacterium tumefaciens和Sphingomonas pseudosanguinis。这两株降解菌能够在15℃,原油浓度3 g/L的条件下,7 d降解率分别达到48.25%和38.56%。为提高降解率,对两株菌进行复配,结果表明以2︰1比例复配时其降解率达到56.49%,高于单菌株降解率。
【Abstract】 In order to screen out the strain that can degrade petroleum hydrocarbon under low temperature,the degradation ability of the strain was explored.Two low-temperature petroleum hydrocarbon degrading bacteria were obtained from the soil contaminated by petroleum for a long time in Karamay oil field through enrichment,isolation and purification.These two strains were identified by morphology,physiology,biochemistry and 16 S rDNA for Agrobacterium Tumefaciens and Sphingomonas Pseudosanguinis.Under the condition of 15 ℃ and 3 g/L crude oil concentration,the degradation rate of these two biodegradable bacteria reached 48.25 % and 38.56 % respectively in 7 days.In order to improve the degradation efficiency,the two strains were compounded,and the results showed that the degradation rate reached 56.49 % when the two strains were compounded in a 2︰1 ratio,which was higher than that of single strain.
