【摘要】 目的探讨STAT3和MMP-2在维吾尔族乳腺癌患者中的表达及其临床意义。方法运用RT-PCR和免疫组化方法检测STAT3、MMP-2蛋白和mRNA在80例维吾尔族乳腺癌和30例正常乳腺组织中的表达。比较两者在不同临床病理学特征乳腺癌组织中表达的差异,分析两者的关系;COX回归分析影响维吾尔族乳腺癌的预后因素。结果乳腺癌组织中STAT3和MMP-2蛋白和mRNA的表达水平明显高于正常乳腺组织(P<0.05);STAT3和MMP-2蛋白在维吾尔族乳腺癌中的表达呈正相关(P<0.05);乳腺癌组织中STAT3、MMP-2蛋白的表达与肿瘤大小、组织学分级、淋巴结转移及临床分期有显著相关性(P<0.05);乳腺癌组织中STAT3、MMP-2 mRNA的相对表达量与淋巴结转移及临床分期有显著相关性(P<0.05)。绘制乳腺癌患者术后60个月生存曲线,STAT3和MMP-2 mRNA低表达组患者生存率明显高于高表达组患者(P<0.05)。结论 STAT3和MMP-2蛋白和mRNA表达高的维吾尔族乳腺癌可能更具有侵袭性,预后更差。STAT3和MMP2在维吾尔族乳腺癌发生、发展过程中可能发挥着重要作用,是乳腺癌患者独立的预后因子。更多还原

【Abstract】 Objective To investigate the expression and clinical significance of STAT3 and MMP-2 protein and mRNA in breast cancer tissues of Uygur population. Methods The expression of STAT3 and MMP-2 was assessed by RT-PCR and immunohistochemistry in 80 breast cancer specimens and 30 normal tissues of Uygur population and compared. COX regression analysis of prognostic factors of Uygur breast cancer. Results The positive expression rate of STAT3 and MMP-2 protein and mRNA in Uygur breast cancer were significantly higher than that in normal tissues(P<0.05). The expression of STAT3 and MMP-2 protein was positively correlated in Uygur breast cancer(P<0.05). In Uygur breast cancer, STAT3 and MMP-2 protein were significantly correlated with tumor size, tissue grade, lymph node metastasis and clinical stage(P<0.05). STAT3 and MMP-2 mRNA were correlated with lymph node metastasis and clinical stage(P<0.05). Survival curves of breast cancer patients after 60 months were drawn. The overall survival of patients with low expression of STAT3 and MMP-2 was significantly higher than that of patients with high expression(P<0.05). Conclusion Uygur breast cancer with positive STAT3 and MMP-2 may be more invasive and have a poor prognosis. STAT3 and MMP-2 may play important roles in the development of Uygur breast cancer, and it is an independent prognostic factor for breast cancer patients.更多还原
