植物生长调节剂及培养条件对大花序桉试管苗玻璃化的影响
Effect of Plant Growth Regulators and Culture Conditions on Vitrification of Eucalyptus Cloeziana During Tissue Culture
【摘要】 【目的】探明植物生长调节剂与培养条件对大花序桉试管苗玻璃化的影响机制,为解决大花序桉组织培养过程中玻璃化现象严重的难题提供一定参考依据。【方法】采用单因素试验设计和正交试验设计,研究植物生长调节剂(6-BA、NAA和IBA)及培养条件(培养基添加剂与培养条件)对大花序桉试管苗玻璃化的影响。【结果】6-BA、蔗糖、光照时间、温度是引起大花序桉试管苗玻璃化现象的4个主要原因;低浓度6-BA(≤0.3mg/L),大花序桉玻璃化率低(≤3.5%);适当提高蔗糖浓度能有效控制大花序桉试管苗玻璃化;增加光照时间、降低培养温度对防治玻璃化有一定作用。【结论】大花序桉最佳培养条件:在改良MS培养基(附加0.3mg/L 6-BA、0.1mg/L NAA、0.15mg/L IBA、30g/L蔗糖、6g/L卡拉胶)的基础上,培养温度20~25℃,每天接受10~12h强度为2 500~4 000lx全光谱光照条件下培养,能有效控制大花序桉试管苗玻璃化率,且增殖系数较高,有效试管苗生长较快。
【Abstract】 【Objective】The influencing mechanism of plant growth regulators and culture conditions towards vitrification of Eucalyptus cloeziana test-tube seedlings is explored,which to some degree provides reference to solve the problem of excessive vitrificaiton during culturing tissue of E.cloeziana.【Method】Single factor experiment design and orthogonal experiment design are used to study the effect of plant growth regulators(6-BA,NAA and IBA)and culture conditions(medium additives and culture conditions)on the vitrification of E.cloezianatest-tube seedlings.【Result】6-BA,sucrose,lighting time and temperature are the main reasons why the vitrification of E.cloezianais too severe.When decreasing concentration of 6-BA (≤0.3 mg/L),the vitrificaiton rate of E.cloeziana decreased as well.When increasing the concentration of sucrose properly,the vitrificaiton of E.cloeziana test-tube seedlings is controlled effectively.Prolonging light time and lowering temperature have certain effect on preventing vitrification.【Conclusion】The best culture condition:on the basis of amending MS culture medium(additional 0.3 mg/L 6-BA,0.1 mg/L NAA,0.15 mg/L IBA,30 g/L sucrose,6 g/L carrageenan),the culture condition of keeping temperature as 20~25℃,receiving full-spectrum light with intensity of2 500~4 000 lx for 10~12 ha day can availably control the vitrificaiton rate of Eucalyptus cloeziana testtube seedlings.The proliferation coefficient is higher meanwhile,and the effective buds in vitro growth faster as well.
