长链非编码RNA MALAT1对肺癌细胞放疗敏感性的作用

【摘要】 目的研究长链非编码RNA (LncRNA)肺腺癌转移相关转录本1 (MALAT1)对肺癌细胞放射治疗敏感性的作用。方法运用实时荧光定量逆转录聚合酶链反应(qRT-PCR)检测肺癌细胞A549中MALAT1的表达,应用慢病毒在A549细胞中干扰或者过表达MALAT1,并通过克隆形成实验检测干扰或者过表达MALAT1后A549细胞放疗敏感性的改变,通过Western blot检测干扰或者过表达MALAT1后A549细胞放疗后DNA损伤标记物γ-h2ax表达的改变。结果通过慢病毒在A549细胞中干扰MALAT1后,A549细胞的放射敏感性增加,放疗后DNA损伤标记物γ-h2ax的表达增加。通过慢病毒在A549细胞中过表达MALAT1后,A549细胞的放射敏感性降低,放疗后DNA损伤标记物γ-h2ax的表达减少。结论调控长链非编码RNA MALAT1可以影响肺癌细胞的放射治疗敏感性,且有可能成为肺癌放疗增敏的新靶点。

【Abstract】 Objective To study the effect of long non-coding RNA MALAT1 on radiosensitivity of lung cancer cells.Methods qRT-PCR was used to detect the expression of MALAT1 in lung cancer cells A549,lentivirus was used to knockdown or overexpress MALAT1 in A549 cells,and clone formation experiment was used to detect the changes in radiosensitivity of A549 cells after knockdown or overexpression of MALAT1,Western blot was used to detect the changes in the expression ofγ-h2 ax,a radiation induced DNA damage marker in A549 cells after knockdown or overexpression of MALAT1.Results After the knockdown of MALAT1 in A549 cells by lentivirus,the radiosensitivity of A549 cells increased,and the expression of DNA damage markerγ-h2 ax increased after radiotherapy.After overexpressing MALAT1 in A549 cells by lentivirus,the radiosensitivity of A549 cells was reduced,and the expression of DNA damage markerγ-h2 ax after radiotherapy was reduced.Conclusion The regulation of long non-coding RNA MALAT1 can affect the radiosensitivity of lung cancer cells,and it may become a new target for lung cancer radiotherapy sensitization.
