鸡粪堆肥高温腐熟菌的分离和鉴定及其初步应用

【摘要】 为了从鸡粪中分离高温菌用于鸡粪在堆肥反应器中的快速腐熟,利用Luria-Bertani(LB)培养基分离鸡粪中的高温腐熟菌并对菌株进行初步分类鉴定,利用解淀粉、解蛋白质、解纤维素和抑菌方法筛选不同腐熟功能和抗菌活性的菌株用于考察鸡粪堆肥过程中促进腐熟减少臭味的效果。结果从鸡粪分离得到YJFJ4、YJFJ6、YJFJ8、YJFJ9和YJFJ12共5株活性菌,经16S rRNA基因序列分析鉴定,分别属于特基拉斯芽孢杆菌、副地衣芽孢杆菌、暹罗芽孢杆菌、地衣芽孢杆菌和枯草芽孢杆菌粪便亚种,它们对淀粉、蛋白质和纤维素均有不同程度的降解能力,对致病菌大肠杆菌、金黄色葡萄球菌和沙门氏菌也有一定的抑菌活性。将这些活性菌株制成复合菌剂用于鸡粪堆肥发酵,能够明显提高腐熟速度、温度,并减少臭味的产生,促进腐熟的完全,实现资源化利用。

【Abstract】 To isolate thermostable strain from the chicken manure for chicken manure speed-up compositing in a composting reactor,the strains in the chicken manure were isolated from the Luria-Bertani medium,and the strains were preliminarily classified and identified. The ability of hydrolyzing starch,degrading protein and cellulose,and antibacterial activity of the strains were studied,and strains with different ripening function and antibacterial activity were used to investigate the effect of promoting compost and reducing odor in chicken manure composting process. Results show that 5 strains of YJFJ4、YJFJ6、YJFJ8、YJFJ9 and YJFJ12 were isolated from the chicken manure and identified as Bacillus tequilensis,B. paralicheniformis,B. siamensis,B. licheniformis and B. subtilis subsp.Stercorisi identified by 16 S rRNA genes sequence analysis. They had different degrees of degradation to starch,protein and cellulose,some antibacterial activity against Escherichia coli,Staphylococcus aureus and Samonella. The mixed strains used in chicken manure composting could obviously increase the maturity speed,temperature,and reduce the production of odor,promote maturity and realize the utilization of resources.
