组蛋白去乙酰化酶在变应性鼻炎鼻黏膜上皮中的表达研究

【摘要】 目的从组织学层面对比研究变应性鼻炎(AR)鼻黏膜上皮和正常鼻黏膜上皮中组蛋白去乙酰化酶(histone deacetylases,HDACs)的表达情况。方法首都医科大学附属北京同仁医院鼻过敏科手术患者中,收集6名对照鼻黏膜和7例AR患者鼻黏膜,应用实时荧光定量PCR和免疫组化染色分别检测HDAC1、HDAC9、SIRT6、SIRT7 mRNA和蛋白的表达情况。结果实时荧光定量PCR结果显示,HDAC1、HDAC9、SIRT6和SIRT7的mRNA在AR鼻黏膜中表达较正常对照组上调,尤其是HDAC1、HDAC9和SIRT7;免疫组化染色显示,AR组中这4种HDACs在鼻黏膜上皮中表达增强。结论 AR鼻黏膜上皮细胞多种HDACs表达上升,可能与AR黏膜屏障功能下降、变应原得以进入体内引发变态反应相关。

【Abstract】 OBJECTIVE To investigate the expression of histone deacetylases(HDACs) on the nasal epithelial cells of allergic rhinitis(AR) and healthy subjects.METHODS 6 normal nasal mucosa samples and 7 AR mucosa samples of patients who had endonasal endoscopic surgery were enrolled.Immunohistochemical staining and real-time fluorescence quantitative PCR was conducted to evaluate the protein and mRNA expression of HDAC1,HDAC9,SIRT6 and SIRT7 respectively.RESULTS Both the immunohistochemical staining and the RT-PCR demonstrated increased expression of HDAC1,HDAC9,SIRT6 and SIRT7 in the nasal epithelial cells of AR patients,especially HDAC1,HDAC9 and SIRT7.CONCLUSION The increased expression of HDACs in the nasal epithelial cells of AR patients might play a role in the dysfunction of nasal epithelial barrier which result in the pathogenesis of AR.
