禽网状内皮组织增生症病毒感染雏鸡血液T细胞数量和细胞因子含量变化

【摘要】 为探索禽网状内皮组织增生症病毒(REV)感染对雏鸡外周血液细胞免疫功能及其相关细胞因子的影响,将60只1日龄SPF雏鸡随机分为REV感染组和未感染REV的对照组,应用流式细胞术结合ELISA等免疫学方法,通过血液T淋巴细胞亚型数量与比值变化,以及其免疫相关因子含量等动态变化的检测,系统地研究REV感染对SPF雏鸡血液细胞免疫功能的影响。结果显示,REV感染SPF雏鸡外周血液CD4~+T淋巴细胞在整个试验期间均明显低于对照雏鸡(P<0.05),而CD8~+T淋巴细胞数量除病毒感染后第7天明显增加(P<0.01)外,其余被检时间点均显著低于对照雏鸡(P<0.05或P<0.01), CD4~+与CD8~+T淋巴细胞比值在病毒感染早期较对照雏鸡明显增加,而病毒感染后期显著低于对照雏鸡,与相应对照雏鸡比较,REV感染SPF雏鸡血液IL-2和IFN-γ含量明显降低(P<0.05),而IL-4和IL-10含量显著上升(P<0.05)。结果表明,REV感染所致的SPF雏鸡血液免疫功能抑制与其降低雏鸡外周血液免疫增强因子含量,增加免疫抑制因子含量及减少CD4~+和CD8~+T淋巴细胞数量密切相关。

【Abstract】 In order to explore the impact of avian reticuloendotheliosis virus(REV) infection on the immune function of peripheral blood cells in chickens, a total of 60 1-day-old SPF chicks were randomly divided into REV infection group and control group.Flow cytometry and ELISA were used to detect changes in the number and proportion of T lymphocyte subtypes, and immune-related cytokines.The results showed that the peripheral blood CD4+ T lymphocytes of REV-infected SPF chicks were significantly lower than that of control chicks during the whole experiment period(P<0.05),and the number of CD8+ T lymphocytes was lower than that of control chicks except for the 7 th day after virus infection, the ratio of CD4+ to CD8+ T lymphocytes was significantly higher than that of control chicks in the early stage of virus infection, and was significantly lower than that of control chicks in the late stage of virus infection, and compared with the corresponding control chicks, the blood IL-2 and IFN-γ levels of REV-infected SPF chicks were significantly decreased(P<0.05),while the levels of IL-4 and IL-10 were significantly increased(P<0.05) The above results indicated that REV infection can reduce the contents of peripheral blood immune enhancing factors and increase the contents of immunosuppressive factors and reduce the numbers of CD4 and CD8 T lymphocytes, resulting in suppression of immune function in chickens.
